OBJECTIVE:To study the anti-aging effect of thymopentin and its anti-aging mechanism at cellular level. METHOD:Mouse skin fibroblasts was studied for its cell culture method and identified from five aspects: cell activity, proliferation, cell morphology, chromosome and species specificity. Then the skin fibroblasts was used as administration carrier to observe the effects of thymopentin on cell proliferation, cell passage, survival days, activity of superoxide dismutase(SOD) and content of malondialdehyde(MDA)in cell. RESULT:Mouse skin fibroblasts are isolated by modified trypsin digestion method, and new methods for cell purification,cell growth, cell passage and cryopreservation are developed and used in the following cell experiments. The survival rate studied by Trypan blue staining technique was found to be over 65% in the primary cells,and was over 95% in the passage cells. Cell proliferation ability demonstrated that the growth of mouse skin fibroblasts were stable and complied with the general trend of cell growth. Cell morphological study showed that the cultured cells possess typical characteristics of fibroblasts. The number of chromosome was 40 thus suggested its murine origin. The positive result of immunohistochemical of vimentin indicated that the cultured cells were fibroblasts. The studies on the anti-aging effect of thymopentin showed that thymopentin significantly promoted the proliferation of cell (P<0.001), increased the number of cell passage (two generations) and prolonged the survival time of cell from 13 days to 36 days,when compared with the blank control group,. It also showed thymopentin improved the total SOD activity in cells (P<0.001), and decreased the total amount of MDA produced (P<0.001). CONCLUSION:Thymopentin delayed the aging process in cellular model of mouse skin fibroblasts since it promoted the proliferation of mouse skin fibroblasts, increased the number of cell passage, prolonged cell survival time,improved the SOD activity and decreased the content of MDA in cells.The skin fibroblast model can therefore be a promising high throughput drug screening model for anti-aging drugs.
