Objective1.To establish the Bleomycin-induced pulmonary fibrosis model in rats;2.To observe the effects of puerarin on the pulmonary fibrosis in rats. 3.To explore the mechanism of the effects of model SD rats by the histopathology, the activity of the SOD, the contents of HYP and MDA.MethodsForty male SD rats were divided into four groups: control group( N=10), model group( N=10), puerarin group( N=10) and glucocorticoids group( N=10), (1)The rats of control group were injected normal sodium with a dose of 5mg/kg in trachea at the beginning of experiment.(2)The rats of model group were injected bleomycin A5 with a dose of 5mg/kg in trachea at the beginning of experiment. Both the control group and the model group were administered intraperitoneal injection of normal sodium with a dose of 1ml/kg.(3)The rats of puerarin group were injected bleomycin A5 with a dose of 5mg/kg in trachea at the beginning of experiment and simultaneously administered intraperitoneal injection of puerarin with a dose of 30mg/kg. (4)The rats of glucocorticoids group were injected bleomycin A5 with a dose of 5mg/kg in trachea at the beginning of experiment and simultaneously administered intraperitoneal injection of glucocorticoids with a dose of 1mg/kg. On the 7th, 28th day after administration,5 rats of each group were sacrificed respectively. On the 28th day,the lungs were harvested for histophological examintion, the lung coefficients and hydroxyproline(HYP) of lung tissues hemogenate were decected. At the same time, the contents of superoxide dismutase(SOD) and malonyldialde- hyde(MDA) were detected respectively.ResultsThe comparison of the lung coeffcients : The lung coeffieients in the model group and in the other group were significantly (P<0.01). Howevert there was no significance between puerarin group and glucocorticoids group(P>0.05). (2) By the HE staining the alveolar inflammation and the Pulmonary fibrosis were more mitigated obviously in puerarin and glucocorticoids groups than model group; after treating for 7 days,pulmonary inflammation cells and infiltration area were more mitigated obviously in puerarin and glucocorticoids groups than in model Group; after treating for 28 days,the pulmonary fibrosis in puerarin and glucocorticoids group was changed lightly and moderately,the collagen fibers in pleural membrane and alveolar membrane were bunehy.(3)The contents of SOD in lung tissues homogenate of puerarin and glucocorticoids groups were higher than model group(P<0.01); but the contents of MDA and HYP in lung tissues homogenate of puerarin and glucocorticoids groups were lower than model group(P<0.01);during each period,the activities of SOD and the contents of MDA and HYP in lung tissues homogenate of control group were consistent basically.Conclusionpuerarin could significantly alleviate the degree of the alveolitis and pulmonary fibrosis,increase activity of SOD in lungs the rats with pulmonary fibrosis, and lower the content of MDA and HYP.
