| Objective:To observe the impact on traditional Chinese medicine Yi Kun ning in the perimenopausal ovarian cytochrome P450 aromatase(P450arom).Methods:Select natural aging rat model to be perimenopausal ovarian function recession.Then they were randomly divided into model group,Yi Kun Ning-group,the control group of western medicine and the control group of younth.Yi Kun ning of Chinese medicine and liviai water were given to Yi Kun ning medicine group and the control group of western medicine.After four weeks,we detect the expression of P450arom mRNA and protein on the ovarians with the in situ hybridization,immunohistochemistry and western blot.At the same time,we detect the estradiol(E2)level with RIA determination.Compared Yi Kun ning group with the other groups to observe the impact on aromatase cytochrome P450 on the of perimenopausal ovarian of Yi Kun ning of Chinese medicine.Results:P450arom mRNA and protein mostly express negatively on ovarians in Yi Kun ning group,compared with the model group,it have the most significant difference(P＜0.01);control group and the difference between Western medicine does not have statistical significance(P＞0.05).Yi Kun ning rats fasting serum estradiol(E2) levels increased,compared with the model group difference was significant(P＜0.01),compared to the control group and western medicine does not have significant differences(P＞0.05),but its E2 levels are still below the young control group,the difference was significant(P＜0.05).Conclusion:Perimenopausal natural aging rats can copy the successful model.Chinese medicine Yi Kun ning would prefer to promote peri-menopausal period of ovarian P450arom mRNA expression.Chinese meidicine Yi Kun ning would prefer to promote peri-menopausal period of ovarian P450arom protein expression. Chinese Yi Kun ning would prefer to raise serum estradiol(E2)level. Yi Kun ning prefer traditional Chinese medicine through the promotion of ovarian P450arom conducive to the expression of estrogen production. |
