Quercitrin(QU) and Ursolic acid(UA) is distributed widely in nature. QU is the major chemical components of Chinese medicine, such as heartleaf houttuynia herb and Chinese taxillus herb. UA is existenced in Chinese medicine, such as glossy privet fruit, Chinese hawthorn and plantaginis. Domestic and international studies show that QU and UA have a central inhibition effect.For finding safe and effective medicines to improve sleep, the effects of QU and UA on sedation and hypnosis were evaluated by following researches. First of all, QU and UA on pentobarbital sodium induced sleep time of mice were observed. Based on this, we study the sedative and hypnotic effects of dose- and time-effect both of them. Meanwhile the effect of QU and UA on the endogenous substances of sleeping was studied.Experimental Result:1.The effect of QU and UA on pentobarbital sodium induced sleep time of mice: QU (50mg/kg) can significantly prolong the sleeping time in mice(P<0.01), UA(50mg/kg) can extend the sleeping time in mice (P<0.01) .2.The study about dose-effect relationship of QU and UA prolong the sleeping time on pentobarbital-induced sleep in mice: QU below 20 mg / kg dose did not show significantly prolonged effect, however, more than 20 mg / kg there was significant dose-effect relationship (P<0.01) ,and more than 40 mg / kg the prolonged effect would not increase. UA at dose of 64 mg / kg has significant synergism of pentobarbital sodium induced sleep time of mice (P<0.01) ,other does showed no significant prolonged effect. Less than 64 mg / kg there was significant dose-effect relationship, but more than 64 mg / kg, prolonged effect was the slow downtrend.3.The study about time-effect relationship of QU and UA prolong the sleeping time on pentobarbital-induced sleep in mice: QU at the dose of 60mg/kg and UA at the dose of 64mg/kg could obvious prolong effect on the sleeping time in mice treated with pentobarbital sodium (55mg/kg, i.p.) 0.25h, 0.5h, 1h later (P<0.05) . QU and UA had the effect treated with pentobarbital sodium 15min later and maintained 1h.4. The effect of QU and UA on the five endogenous substances :QU can significantly elevated pieces and magnitude of SP positive cell in rat cortex (P<0.01), pieces and magnitude of SP and VIP positive cells in cornu ammonis (P<0.05), pieces and magnitude of GABA positive cells in cortical area (P < 0.01); QU can decrease Glu positive cells in cortical area (P<0.01). QU has no significant effect of CCK in various brain regions, and has no effect of the five substances in Hypothalamus area.UA can increase pieces of SP positive cells in cornu ammonis (P<0.05), significantly increase pieces and magnitude of VIP positive cells in Hypothalamus ( P<0.01), pieces of CKK positive cells in cortical area (P<0.05) and intensity (P<0.01); GABA positive cells in cortical area, pieces( P<0.05) and magnitude(P<0.01) of GABA positive cells in cornu ammonis; UA can significantly decrease pieces(P < 0.05)and magnitude( P < 0.01) of Glu positive cells in cortical area, pieces(P<0.05)and magnitude(P <0.05) of Glu in cornu ammonis.The conclusions above provided an experimental basis for further study of QU and UA on the sedative and hypnotic effects meanwhile they were laid the foundation for improve sleep of new drug research.
