Study on regeneration of spinal cord is an important task in neurosciences. It began in the 1920's but was interrupted for several decades. The research in this field did not start again in 1980's until Aquago did his outstanding work which told the world that the CNS, including spinal cord, could regenerate, if it was provided with enough necessary substances, for example neurotrophic factors (NTFs). So, NTFs are the crucial elements in the regeneration of spinal cord. The aims of this experiment were to study neurotrophic effects of those neurotrophic substances secreted by spinal cord itself on spinal cord neusons, especially on non-motor neurons and to isolate and purify those substances.Crude extracts were prepared from 4 month and 7 month fetus respectively, and were further divided into three fractions, < 10KD, 10-30KD and > 30KD with centricon (Millipore). Spinal cord cells used for culture in vitro came from embryonic 12 day (E12) mouse spinal cord, which served as a model for studying the neurtrophic effects of the endogenous substance of spinal cord.
