| 0bjective:To investigate the role of Danshensu on transforming growth factor (TGFβ1) stimulated signal transduction in rat hepatic stellate cells(HSCs).Method:The mice HSCs were isolated and cultured for the following study.MTT colorimetric assay was used to detect proliferation of HSC with different concentration of Danshensu.α-smooth muscle actin(α-SM A)expression was analyzed by immunocytochemical staining. The mRNA level of Smad2,3,7 were measured with RT-PCRResults:(1) Danshensu in the concentration of 0.0625~1mmol/L could prevent the proliferation of HSC in a dose-dependent manner(P<0.05);Danshensu also had an inhibiting effect on the proliferation of HSC induced by TGFβ1 in a dose-dependent manner (P<0.05).(2)The concentration of 0.25 mmol/ L Danshensu could downregulateα-SMA protein expression of HSC,with or without stimulation of TGFβ1(P<0.05),and the activation of HSC was inhibited also.(3) TGFβ1 increased the mRNA level of Smad2 , 3 , 7 in HSC;Danshensu could downregulate the mRNA level of Smad2,3 and upregulate the mRNA level of Smad7 in HSC induced by TGFβ1 (P<0.05).Conclusion:Danshensu could directly or indirectly restore the normal transmembrane signal of TGFβ1 through upregulation of Smad7 mRNA expression and downregulation of Smad2,Smad3 mRNA expression in HSC. |
