Objective:To explore the dependability between the aging variation of expression of peroxisome proliferator-activated receptorγ(PPARγ)and core binding factora1(Cbfa1)and aging fracture healing impairment, we studied the expression of PPARγand Cbfa1 in aging fracture healing.Methods:Twelve male Sprague-Dawley(SD)Rats were divided into 2 groups,the young group(n=6)(3-month-old)and the aged group (n=6)(23-month-old).All the rats received osteotomies on the left tibia and placement of special external distractive fixators.The distraction was taken from the following day of the operation for 14 days with 0.2mm, Bid(0.2mm/d).Animals were sacrificed on the 15th days.The lengthened left tibea were harvested and analyzed using imageological,histological and the Reverse Transcription- Polymerase Chain Reaction(RT-PCR) techniques.All the rats were included into the analysis for the results.Results:More new bone formation existed between the broken ends of the fracture and bone formation was obvious in young group imageologically.Great number of new bone formation and significant membrane bone formation were seen in the distracted area in young group in histological technique.In aged group,osteotylus were rear and only fibrous joint were seen.In RT-PCR examination,expression of Cbfa1 was higher in young group(P<0.05),and expression of PPARγwas higher in aged group(P<0.05). Conclusion:The ability for fracture healing decreased and the expression of PPARγand Cbfa1 changed,demonstrating the relation between the expression of the two factors and the aging fracture healing impairment.
