Objective:An animal model of mice skin photoaging was established,then treated with IPL(intense pulsed light,IPL).we evaluating comprahensively and understand the function of IPL from different aspects by comparing the changes on biochemical index,organization sturctrue,pathomorphology and cytokines in skin before and after treated with IPL.Methods:①Stablished a skin photoaged model combined Ultraviolet A and Ultraviolet B;②Grouping:Negative control group and model group were established,IPL treated group(1 week and 3 weeks after IPL teated)and un-treated group(1 week and 3 weeks after un-teated by IPL)were deveided from the model,when model was succeed;③Detecting biochemical index:The activity of SOD(Superoxide Dismatase,SOD)as well as the contents of(MDA Malcic Dialdehyde,MDA)and HYP(Malcic Dialdehyde,MDA)in the skin before and afeter IPL treatment were examined by Ultraviolet spectrophotometer;④tectology observation:the changes of the organization structure,collagen fiber and elastic fiber of mice skin were detected by HE staining and orcein staining respectively;⑤Detecting cytokines:the expression and distribution of NF-kB,VEGF and TNF-a pre and post IPL treatment in the mice skin were examined by S-P staining.Results1.Visual inspection:significant photoaging-charactersitic in the mice of raodel group appeared;2.Evaluation of biological markers:there was a statistical significance between the light absorption value of SOD,MDA,HYP and the normal group(P<0.05);after the IPL treatment,there was little changes happened on the light absorption of SOD,MDA,no statistical significance(P>0.05),only HYP had a statistical significance comparing with these un-treated groups(P<0.05).3.Observing the morphology of pathological slices:Normal group:The epidermis was very thin and have few cell layer, even be monolayer,the arrangement of collagen fiber in dermis was sparse and scattered,which was tenuity and pink;Model group:The epidermis was thickened significantly, accompanying with hyperkeratinization or parakeratosis,and the volume of epidermis cells was enlarged with hyperchromatin,the collagen in the shallow dermis layer thickened,homogenized and high stained.IPL-treated groups:The epidermis of IPL-treated groups graduatly became thinner and un-treated groups;the collagen layer was thickened in the shallow dermis layer and tissue arrangement was tightened;Un-treated groups:The epidermis of un-treated groups graduatly became thinner,but there was little change in the dermis;4.The measurement of skin thickness:The thickness values of epidermis in the model group mice were significantly higher than the normal group mice,and the difference had statistical significance(P<0.05);The thickness values of dermis in the model group mice were significantly lower than the normal group mice,and the difference also had statistical significance(P<0.05).After one-week's IPL treatment,the thickness values of epidermis were slightly higher than the one-week un-treated group,but there was no statistical difference(P>0.05,the thickness value of dermis between these two groups was no statistical difference(P>0.05);the thickness values of epidermis between the three-week's IPL treatment group and three-week's un-treated group was no statistical difference(P>0.05),the thickness value of dermis between these two groups had statistical significance(P<0.05)5.Elastic fiber staining:Made the elastic fibers to be brownish black by orcein staining,then we observed that more,thicken and feformed elastic fibers,after the IPL treatment,the elastic fibers might decreased gradually.6.The area of elastic fibers:The difference between the model group and the normal control group had a statistical significance (P<0.05);the difference between the IPL treated 3w group and the post-modeling 3w group also had a statistical significance(P<0.05).7.Immunohistochemical staining:①The expression of NF-κB:the difference between the model group and the normal group was notable,and it had a statistical significance (P<0.05);there was a statistical difference(P<0.05)between the IPL treated 1w group and the post-modeling 1w group;there was no statistical significance between the post-modeling 3w group and the IPL treated 3w group(P>0.05);②The expression of TNF-a:the difference between the model group and the normal group was notable and it had a statistical significance(P<0.05);there was a statistical difference(P<0.05)between the IPL treated 1w group and the post-modeling 1w group;and there was no statistical significance between the post-modeling 3w group and the IPL treated 1w group(P>0.05);③The expreesion of VEGF:the difference between the model group and the normal group was notable and it had a statistical significance(P<0.01);there was a statistical difference(P<0.05)between the IPL treated 1w group and the post-modeling 1w group;there was no statistical significance between the post-modeling 3w group and the IPL treated 3w group(P>0.05).Conclusion①The mice,which had been irradiated for 8 weeks by UVA combining with UVB,had the basis for the clinical and pathological diagnosis of the model of skin photoaging.②The IPL has tiny stimulation for the epidermis,the thickening of epidermis caused by the IPL was not long lasting in the morphopathological changes.③The IPL acted mainly on the dermis.④Significant correlation was found between the expression of NF-κB,TNF-a,VEGF and the skin photoaging.⑤The re-expression of NF-κB,TNF-a,VEGF after the IPL treatment suggested that they might participated in the thermal damage repairing process induced by the IPL,and a temporal correlation might exist among them.
