Objectivesâ‘ To establish a skin aging model for studying the process of skin aging and potential compounds against skin aging.â‘¡To establish the mechanism of action for skin aging with intense pulsed light.Methodsâ‘ With D-galactos combined with UVA and UVB irradiation on the dorsal skin of mice,a model was established resembling to human skin aging.Then the morphologic alteration of dorsal skins was observed under microscope,and analyzed by computer systems.The biochemical indicator and the change of apoptosis-associated gene proteins Bax,Bcl-2 were monitored.â‘¡The model were irradiated by the IPL system in vivo under certain wavelengths with different energy densities,continuously irradiated two times,interval two weeks.Then the morphologic alteration of dorsal skin was observed under microscope,and analyzed by computer systems. The biochemical indicator and the change of apoptosis- associated gene proteins Bax,Bcl-2 were monitored.Resultsâ‘ Compared with D-galactose and UV groups,the model group indicted that the epidermis was increased,dermis was significantly reduced, elastic fibers were accumulated and tangled.The activity of SOD and content of hydroproline were statistically decreased,whereas the content of MDA were remarkably increased.â‘¡Compared with D-galactose and UV groups,the model group indicated that the apoptosis-associated gene protein Bax was increased, while the apoptosis-associated gene protein Bcl-2 was reduced.â‘¢Compared with the model control group,the model irradiated by the IPL system in vivo indicated that the dermis was significantly increased,collagen fibers were raised.The activity of SOD and content of hydroproline were statistically increased,whereas the content of MDA were remarkably decreased.â‘£Compared with the model control group,the model irradiated by the IPL system in vivo indicated that the apoptosis-associated genes protein Bcl-2 was increased.Conclusionâ‘ D-galactose combined with UV can cause skin aging remarkably and rapidly.It is an ideal tool to study skin aging.â‘¡The apoptosis-associated genes protein Bax,Bcl-2 may play an initial role in the pathogenesis of the skin aging.â‘¢IPL is an effective method for D-galactose and UV group,the mechanism of which may improve the balance between oxidation and antioxygen, and recover oxydasic active.â‘£IPL is an effective method for D-galactose and UV group,the mechanism of which may improve the apotosis of skin and raise apoptosis -associated inhibiting gene protein Bcl-2.
