Objectives:To elucidate the mechanism of drug-resistance in nasopharyngeal carcinoma cells,establishment of paclitaxel-resistance cell lines of nasopharyngeal carcinoma was carried out and relationship between drug-sensitivity,-resistance and FOLR1 expression level was studied.Methods:1.The human nasopharyngeal carcinoma HNE-2 cells growed in medium containing paclitaxol,large dose paclitaxel attacking the cells then low dose paclitaxel maintaining in the cells,to obtain the resistance lines which named HNE-2/Taxol.2.The colony formation assay was repeatly carried out to detect the half inhibiting concentration(IC50)of the human nasopharyngeal carcinoma CNE-1,CNE-2,HNE-2,and determined the half inhibiting concentration(IC50)and the resistance index of paclitaxel-resistance nasopharyngeal carcinoma cells(HNE-2/Taxol);3.Semi-quantitative RT-PCR was carried out to assess mRNA expression level of FOLR1 in the human nasopharyngeal carcinoma CNE-1,CNE-2,HNE-2 and in the paclitaxel-resistance nasopharyngeal carcinoma cells(HNE-2/Taxol).Results:1.The paclitaxel-resistance nasopharyngeal carcinoma cell lines had been established.The colony formation assay showed that the IC50 value of HNE2/T5 and HNE2/T10 was,respectively,1.7 and 5.23ng/ml. the resistance index was,respectively,1.42±0.15,4.36±0.21;2.The IC50 value of CNE-2,CNE-1,HNE-2 was,respectively, 5.02ng/ml,0.86ng/ml,1.20ng/ml;3.FOLR1 expression level in parental nasopharyngeal carcinoma lines negatively co-related to the drug-sensitivity of Paclitaxel.The correlation coefficient was 0.9923(p<0.05).4.FOLR1 expression level in drug-resistant nasopharyngeal carcinoma lines positively co-related to the drug-resistance of taxol. The correlation coefficient was 0.9944(p<0.05). Conclusion:1.A paclitaxel-resistant human nasopharyngeal carcinoma cell line(HNE-2/Taxol)was established,It is an usable model for exploring the mechanisms of acquired resistance and paclitaxel-resistance.2.FOLR1 expression level in parental nasopharyngeal carcinoma lines negatively co-related to the drug-sensitivity of taxol.3.FOLR1 expression level in drug-resistant nasopharyngeal carcinoma lines positively co-related to the drug-resistance of taxol.4.FOLR1 might be an important target molecule for the reversion of paclitaxel-resistance phenotype in nasopharyngeal carcinoma cell.
