Objective:To establish a reliable method to save the tissue culture cells,and analysis the expression profiling of cytochrome P450(P450)in paclitaxel-resistance nasopharyngeal carcinoma cell lines(CNE1/Taxol) and wild-type(CNE1).Method:1.The half-maximal inhibition concentration(IC50)and the resistant index of cell line to paclitaxel were determined by colony formation assay;2.Cells were frozen by utilizing fast-frozen and slowly-frozen method,and cell survival rates were observed by cell cytometer after cell recovery.2.The expression of p450 family members were detected in nasopharyngeal carcinoma cells by utilizing cDNA microarray technique with the Affymetrix Human Genome U133 Plus 2.0 GeneChip Arrays;3.The differential gene expression of P450 members were analysized comparing CNE1/Taxol with CNE1 by GCOS and Dchip.Result:1.The cell survival rate was much higher in fast-frozen cells than in slowly-frozen cells.2.The results of genechip analysis showed nine members were expressed in nasopharyngeal carcinoma cells in fifty-three members of p450 family.Five genes(CYP1A1,CYP2R1, CYP2U1,CYP3A5,CYP51A1)showed a significantly higher expression in CNE1/Taxol than in CNE1,and expression of CYP1A1 and CYP2R1 was detected to be continiously increased in CNE1/Taxol cells by the treatment of paclitaxol.Conclusion:1.The fast-frozen method is a better cell frozen means to save the cells.2.CYP1A1 and CYP2R1 of the p450 family members may be related to the paclitaxel-resistant phenotype in human nasopharyngeal carcinoma cells.
