Purpose: To discuss the effect of repairing segmental bone defect with the composite of heterogenic deproteinised cancellous bone,bone morphogenetic protein(BMP)and fibrinin.Methods: Bone defects were created in 45 New Zealand rabbits on the bilateral Radius and were treated with three kinds of implantations: A, Composites of heterogenic deproteinised cancellous bone , BMP and fibrin;B, heterogenic deproteinised cancellous bone; C, nothings control. The defects were observed through entgenograph and optical microscope observing, at 2, 4, 8, 12,16 weeks.Results: The defects treat with A implantation regenerated much more new bone and bridged earlier than the defects treated with B implantation and were repaired satisfyingly 16 weeks after operation. The defects with B and C implantation couldn't get osseous tissue healing.Conclusion:1. The composite of caprine deproteinised cancellous bone and fibrinin loaded with bone morphogenetic protein(BMP) displayed fine bone formation activity , did not show conspicuous immunological rejection in muscle bag experiment of mouse. 2. It was easy,cheap and deduplicative to erect experimental animal model of rabbit radial defect, which could be used in the experiment of repairing segmental bone defect.3. As a carrier of BMP, fibrin could enhance adsorbability of BMP in caprine cancellous bone, delayed its release, ensured local effective concentration;the composite had osteoconduction and osteoindution, obviously improved bone formation and reconstruction of segmental bone defect, and bone formation of the composite was also better than the simple carrier.4.The experiment confirmed feasibility of heterogenic cancellous bone combined with BMP and fibrinin, offered to foundational knowledge of the theory of following clinical usage.
