objective:To study the relationship between leptin and enhanced osteogenesis after brain injury by investigating the expression of leptin in callus and the level of leptin in serum of rats with brain injury and femoral fracture.Methods:64 male SD rats were randomly divided into 4 groups: normal control,brain injury group,fracture group,fracture with brain injury group(n=16 per group).Fracture and brain injury models were established.The rats were sacrificed 2,4,8and12weeks respectively after operation,and then the leptin concentration in serum was detected by rat leptin RIT kit and the callus was observed in the X-ray film and volume was measured. The callus was also examed with immunohistochemical staining.Results:The serum levels of leptin in the brain injury group,fracture group or fracture with brain injury group significantly increased as compared with that in control group at the 2nd week after operation(P<0.05),The concentration of the leptin in serum of the fracture with brain injury group was higher than that of brain injury or fracture group 4,8weeks after operation(P<0.05)and declined from 8rd week;at the 12 rd week after operation the difference in above four groups was not statistically significant.(P>0.05).The mean percentage of positive cells in callus of the fracture with brain injury group was higher than that of the fracture group(P<0.05).Callus formation of the fracture with brain injury group were larger and formed more quickly than those of the fracture group(P<0.05).Conclusion:The femoral fracture recovers rapidly and leptin in the callus expresses highly in fracture with brain injury group,which suggests that leptin could yield the healing of the fracture.
