Objective: To investigate the effects of oxidized Low Density Lipoprotein (OX-LDL) on the expression of IL-l0 and MIP-1αin cultured human monocytes from rheumatoid arthritis(RA), and discuss it's role in chronic synovitis and cardiovascular diseases of RA pathogenesis.Methods: Monocytes were isolated from peripheral blood of RA patients with Ficoll-Hypaque, then incubated in RPMI1640 with different amounts of OX-LDL .The contents of IL-l0 and MIP-1αwere evaluated by ELISA in 24h respectively. With RT-PCR, studied the effects of OX-LDL on the expression of cytokines including IL-l0 and MIP-1αin the human peripheral blood monocytes.Results: Monocytes were increased the expression of MIP-1αafter incubating with OX-LDL (10~30μg·ml-1 ) for 24h(P<0.05),and the expression of IL-l0 was reduced. In RA and normal group, the mRNA expression of MIP-1αwas increased (P<0.05). The mRNA expression of IL-l0 was reduced in RA group, but increased in normal group.Conclusions: IL-l0 and MIP-1αinvolve in the formation of chronic synovitis and atherosclerosis. It is an important mechanism that the high expression of MIP-1αand low expression of IL-10 induced by OX-LDL may be contributed to the formation of chronic synovitis and cardiovascular complications of RA pathogenesis.
