Objective: Tumor necrosis factor-alpha(TNF-α) and Interleukin-6(IL-6) are involved in the pathogenetic mechanisms responsible for gestational diabetes mellitus(GDM). TNF-αand IL-6 plasma levels and function may be genetically influenced. We sought to evaluate allelic frequency of the TNF-αand IL-6 in Han ethnicity of China and to investigate whether there is a relativity of Polymorphisms of the TNF-α(G-308A) and IL-6 (C-174G) with pathogenesis of gestational diabetes mellitus.Methods: 90 pregnant women(including 30 gestational diabetes mellitus,30 impaired glucose tolerances and 30 normal glucose tolerances) were recruited. Genotyping for the TNF-α(G-308A) and IL-6 (C-174G) polymorphism was carried out across using Polymerase Chain Reactionrestriction-fragment length polymorphism (PCR-RFLP). The measurement data was analyzed statistically by t-test and enumeration data by chi-square test.Results:1,There was a significant increase of the GA+AA genotype of of TNF-αpromoter -308 gene in GDM group compared with NGT group(73.33% vs 36.67%, P<0.01) and increase of A alleles frequency was obtained in GDM group compared with NGT group(41.67% vs 18.33%, P<0.01).In IGT group, there was a significant increase of the GA+AA genotype and A alleles frequency of TNF-αpromoter -308 gene(60.00%/31.67%, P<0.05).(2)No significant difference was found about the three genotypes among different groups(P>0.05).2,There was a significant increase of BMI in the different genotypes, including the TNF-α(-308G->A) and IL-6 (-174G->C).3,Subjects homozygous for the A of the G-308A TNF-αpolymorphism allele had higher WHR than subjects homozygous for the G(P<0.001), and subjects for GG(P<0.01). Subjects homozygous for the C of the G-174C IL-6 polymorphism allele had higher WHR than subjects homozygous for the G(P<0.05).Conclusion: Polymorphisms of the TNF-α(G-308A) gene was found to have a significant association with gestational diabetes mellitus. There was no significant association between gestational diabetes mellitus and IL-6 (C-174G) genetic polymorphism, which was considered as a dangerous factor to GDM. Subjects homozygous for the A of the G-308A TNF-αpolymorphism allele and subjects homozygous for the C of the G-174C IL-6 polymorphism allele had higher WHR and BMI than subjects homozygous for the G of TNF-αand IL-6, respectively.
