Burn is an emergency disease. The degree of burn will bring the different harm to the body. Burns are difficult to treat, wounds with complex local and systemic pathology and high mortality, that often heal slowly. The syndrome is the main reason of deadly. So finding a cheaper and effective drug is very promising.The course of wound's concrescence is very complicated. Many growth-factors are involved in the course. And the growth-factors are very important factors in the healing. In the literatures show that mRNA of the c-fos,c-myc and bFGF increases in the early of the burns. Even though the principle of protein is not same as the mRNA's, the trend of protein is same as the mRNA's. The others reported that the express of PDGF ,PDGFR and EGFR are increased in the early stage. It is good for the treating that giving the required growth-factors.Now the heparin is the most common medicine ,which is used in the animal or clinical experiment. And then many articles discussed the mechanism of the using heparin to treat burns. The mechanism includes that releasing the condition of burns, mitigating the infections, prevention the syndrome. But it is not discussed that the change of the growth-factors in the wound.The aim of this study was to investigate the mechanism of the change of the growth-factors in the wound healing by the heparin. The study used the method of molecule biology , found the principle of change of the growth-factors.Aim: the study was to investigate the change of the growth-factors in the wound by treating to heparin in the early stage. Comparing the control group with the experiment group, find out the difference in the both groups.Methods : Using the methods of molecular biology. Taking the tissues in the control and experiment groups , extracting and purification the protein in the tissues. By using immobilized pH gradient two-dimensional electrophoresis, we can gain the map of the all proteins. By using the PDQuest , we can analyse the difference in the two teams. By the Western-Blotting, we can find the change of PDGFR-βin different group.Result: (1) By using immobilized pH gradient two-dimensional electrophoresis, we found that the protein spots of control group and heparin-treated group had many difference. The protein spots of experiment group were darker than the control group's.(2) By using the Western-Blot, we found that the expression of PDGFR was increased in the wound tissue. And the experiment group was higher than the control.Conclusion: The mice in the heparin-treat group had a short recovery time. By the molecular biology technology, we found that the expression was higher in the heparin-treat group. These results indicate the foundation for further study on treating burns by heparin.
