Expresson Of Survivin And VEGF In Breast Carcinoma And Correlation With Prognosis

Objective: Through examining the expression of Survivin and VEGF in the mammary gland benign tumor and the breast carcinoma organizes, analyzing their reciprocity and the prognosis relevance with the union integrity clinical material, to provide certain theory basis for breast cancer patient's diagnosis and the prognosis judgment.Methods: Expression of Survivin and VEGF were tested by immunohistochemical method S-P in 17 cases of mammary gland benign tumor and 73 cases of breast carcinoma. The expression of Survivin and VEGF together with lymph node metastasis, tumor size, histology graduation, age, clinical stage, hormone acceptor situation and survivial time were analyzed statistically. The relevance between Survivin and VEGF was analyzed.Results:1.Expression of Survivin and VEGF in breast carcinoma and benign tumor: Survivin predominantly localized to the cytoplasm and the cell nucleus, VEGF was mainly expressed in the cytoplasm. The expression rate of Survivin was 78.1%(57/73) in breast carcinoma that was obviously higher than 23.5%(4/17) in benign tumor. Statistical significance can be found between them (P<0.05); The expression rate of VEGF was 83.6%(61/73) in breast carcinoma that was obviously higher than 40.1%(8/17) in benign tumor. Statistical significance can be found between them (P<0.05).2.Correlation between Survivin and the clinical parameters of breast carcinoma:The expression rate of Survivin was 90.5%(38/42) in the lymph node metastasis that was obviously higher than 61.3%(19/31) in the non-lymph node metastasis group. Statistical significance can be found between them (P<0.05). The expression rate of Survivin was 85.3%(52/61) in the clinical mid and late stage group (II issue, III issue) that was obviously higher than 41.7%(5/12) in the clinical early group (I issue). Statistical significance can be found between them (P<0.05).3.Correlation between VEGF and the clinical parameters of breast carcinoma: There are not significance difference between the expression of VEGF in breast cancer and the age, sex, the menstruation condition, lymph node metastasis and ER, PR, HER-2. The expression rate of VEGF was 94.0%(19/31) in the tumor >2cm group that was obviously higher than 60.9%(14/23) in the tumor≤2cm group. Statistical significance can be found between them (P<0.05). The expression rate of VEGF was 92.1%(58/63) in the clinical mid and late stage group (II issue, III issue) that was obviously higher than 41.7%(5/12) in the clinical early group (I issue). Statistical significance can be found between them (P<0.05).4.Correlations between Survivin and VEGF in breast cancer: There were positive correlations between Survivin and VEGF (P<0.05). The correlation coefficient was 0.449.5.The relationship between survival time of breast cancer patient and the expression of Survivin and VEGF: The five-year total survival rate was 64.6%(42/65), mortality rate was 35.4%(23/65). And the five-year survival rate was 46.9%(15/32) in excessive expression (2+ and 3+) of Survivin and 81.8%(27/33) in low and negative expression (1+ and -) of Survivin. There were statistics significance between the difference (P<0.05). The five-year survival rate was 62.8%(32/51) in excessive expression (2+ and 3+) of VEGF and 71.4%(10/14) in low and negative expression (1+ and -) of VEGF. There were no statistics significance between the difference (P<0.05).Conclusion:1.Survivin and VEGF were low expressed in benign tumor and high expressed in breast cancer.2.Survivin expression correlated in the lymph node metastasis and clinical stages and had no correlation with age, sex, menstruation condition, tumor size and ER, PR, HER-2. 3.VEGF expression correlated in the tumor size and clinical stages and had no correlation with age, sex, menstruation condition, lymph node metastasis and ER, PR, HER-2.4.There was high correlation between the expression of Survivin and VEGF. It was prompted that Survivin possibly participated in the tumor vascularization.5. The excessive expression of Survivin was associated with the survival rate in breast cancer whereas VEGF was negative.
