Objective:ICAM-1 play a important role in embryo development and differentiation, normal tissue maintenance, inflammation and immunity response, cut respire, cruor, thrombus formation, tumor transfer. In this paper we detect the location of ICAM-1 by immunohistochemistry and the expression level of ICAM-1 mRNA in clone cancer tissue and normal tissue to discuss the application of ICAM-1 in clinical diagnosis.Materials and Methods:Sample:There are 32 colonic cancer biopsies including 20 high differentiation and 12 low differentiation. 19 normal tissue biopsies also obtained from routine colonoscopy.Methods:ICAM-1 monoclonal antibody was used for the immunohistchemistry . mRNA was extracted for the tissues and hybridized by the labeled ICAM-1 cDNA probe. The expression level of mRNA was determined by laser densitometry.Data analysis:The data from this experiment was analyzed using the Spss 10.0 system. t-test was used for statistical analysis, with p<0.05 taken as the level of significance.Result: The expressions of ICAM- 1 in colon cancer tissues were significantly higher than those in normal control group ( P<0.05). mRNA northern hybridization analysis show that it is 2.1 fold in colon cancer than in normal tissues.Conclusion: The expression of ICAM-1 has a significant difference between the colon caner and normal tissues. ICAM-1 can be used as a biomarker in the colon cancer diagnosis.
