| Psoriasis is a common, chronic, inflammatory, benign hyper- proliferation disease. It is stubborn ,recurrent and hard to treat. There have not perfect treatment on psoriasis up to now. Recently, Some traditional herb Chinese medicine has been well used on treatment of psoriasis for a long time in our country , though their treatment mechanism has not been known clearly till now. Cooling blood and activating blood circulation formula which integrated traditional Chinese medical theories and experimental formulas was developed following the principles of therapy including cooling blood, activating blood circulation , expelling wind-evil and regulating diuresis. The therapeutic effects of the formula on psoriasis have been tested by animal experiments and clinical studies. And no obvious side effects were observed. Huangqi is one of the major components of Cooling blood and activating blood circulation formula. And Huangqi injection which is extracted from Huangqi herb has been used in psoriatic treatment widely.Objective: In this study, the therapeutic effects of Cooling blood and activating blood circulation formula on psoriasis were compared. The therapeutic effects of Huangqi injection were also observed when combined with Cooling blood and activating blood circulation formula. And the effects of Cooling blood and activating blood circulation formula on proliferation ,apoptosis and in keratinocyte HaCaT were investigated. In this study, we investigated the effects and effective concentration of the traditional Chinese medicine, Cooling blood and activating blood circulation formula, apoptosis in keratinocyte HaCaT. The major purpose of the study is to explore the mechanism of Cooling blood and activating blood circulation formula in antipsoriatic .Methods:(1) Clinical study: 165 patients took cooling blood and activating blood circulation formula for 2 to 4 months. At the same time, some of the patients in were treated with Huangqi injection. Therapeutic effects were determined with PASI scores and comparing the effects. The deference of effective rate between patients treated with Huang-qi injection and not was statistically analyzed.(2) Experimental study: Cultured HaCaT cells in vitro were exposed to Cooling blood and activating blood circulation formula at different concentrations. Inhibition on cell proliferation was measured by MTT assay. Morphological changes were observed under inverted microscope and teansmission electron microscope. Cell cycle and apoptosis were analyzed by flow cytometry.Results:(1) The effective rate successively was 64.2% respectively. And those patients with Huang-qi injection got better therapeutic effects.(2) Cooling blood and activating blood circulation formula and Huang-qi injection exerted notable inhibitory effect on proliferation of HaCaT cells. After treated by Cooling blood and activating blood circulation formula for 24,48,72 hours, the IC50 was 155.83mg/ml, 71.57 mg/ml, 41.27mg/ml;After treated by Huang-qi injection for 24,48,72 hours, the IC50 was 341.0 mg /ml, 252.2 mg /ml, 194.3mg /ml respectively. The inhibition revealed time and dose dependence in effective concentration range.(3) Cell morphological changes and Flow cytometry analysis demonstrated that Cooling blood and activating blood circulation formula could arrest HaCaT cells in G2/M phase and induce apoptosis in a concentration-dependent manner. The results of flow cytometry analysis demonstrated that Huangqi could arrest HaCaT cell in G1 phase and induce apoptosis. The percentage of cells in G1 phage of samples treated was more than negative controlled sample, and that of cells in S/G2 phage were less. Observation of the treated cells under transmission electron microscope showed characteristic opoptotic morphological changes. Conclusion:(1) Cooling blood and activating blood circulation formula is effective on treatment of psoriasis. Huangqi can increase efficiency on psoriasis while combined with the drug above.(2) Cooling blood and activating blood circulation formula can inhibit HaCaT cells proliferation in time and dose dependent manner. They can interfere with HaCaT cell cycle and arrest cells in G1 phage, so that the cells can not get in G/M phage and proliferate. Also Cooling blood and activating blood circulation formula can induce HaCaT cell apoptosis, suggesting that their antipsoriatic mechanism may be relevant to these effects...
