Objective:To investigate the effect of TAP on the diabetic peripheral neuropathy and aorta in the experimental diabetes rats and its mechanisms.Methods:(1)Diabetes Mellitus rats were induced by Streptozotocin(STZ).The diabetic rats are assured if the serum glucose≥16.7mM,and are divided ramdomly into seven group:Diabetic Model,TAP group(390,195,97.5 mg·kg-1,ig),T group (195 mg·kg-1,ig),P group(195 mg·kg-1,ig)and CaD group(100 mg·kg-1,ig),meanwhile,setting the normal rats as the control group.We observed the influence of the complex prescription TAP and its two composed simple recipe T and P on diabetic rats serum glucose,body weight(BW),diet volume,water volnume and urine volume.(2)After 24w administration of TAP,the superoxide dismutase(SOD),malondialdehyde(MDA),glutathione peroxidase(GSH-Px),blood lipid,nitrogen monoxidum(NO)and fructosamine(FMN)in serum and the urinary albumin(UALB) are protected;(3)The nervous conduction velocity of the right sciatic nerve of the diabetic rats is measured.Observe the structure and ultrastructure by the light and electron microscope,meanwhile,analyzing the area and density of myeline sheath of sciatic nerve using the soft of Imageing Analyzing system.Protect the expression of NGF and VEGF in the sciatic nerve using immunohistochemistry,and analyzing the positive cells numbers and the positive intensity using the Imaging analyzing soft mentioned above.(3)After 24w administration of TAP,The structure and ultrastructure of the aorta are observed using the light and electron microscope.The VEGF protein expression is measured by the immunohistochemistry,and the positive cells and positive intensity are analyzing througth the soft described above.Results:(1)TAP can decrease serum glucose,diet volume,water volume and urine volumem meanwhile increase the BW(P<0.05 vs the DM group).(2)TAP can increase the level of SOD,GSH-Px,NO and decrease the content of MDA,TG,TC,LDL-C,GSP in serum and UALB in diabetic rats(P<0.05 vs the DM group). (3)Both the positive cells number and positive intensity of the VEGF and NGF expressed in the sciatic nerve increased obviously compared with the DM group seen from the result of immuohistochemistry(P<0.05),meanwhile,the area and density of the myeline sheath are also increased siginificantly for the TAP group when compared with the DM group(P<0.05).Seen form the light and electron microscope,we observe that the TAP group can lessen the morphology damaging.(4)The TAP has protective effects to the diabetic arota.The morphology damaging of the diabetic arota is lessened when compared with the DM group which show the amotio of the endothelium cells, the migration of vascular smooth muscle cells(VSMC)to the The subendothelial space, the disorder of the VSMCs arrangement in the tunica media and so on.From the results of immunohistochemistry of the arota we can see that both the positive cells number and positive intensity of VEGF decreased obviously than the DM group.T and P group can also produce protective effects compared with the DM group,although their effects is inferior to TAP LOW group.Conclusion:(1)TAP can decrease the diet volume,drinking water volume and the urine volume,meanwhile increase the BW of the diabetic rats obviously.(2)TAP can decrease the serum glucose,lessen the blood lipid disorder and improve siginificantly the antioxidant ability.(3)TAP show an excellent protective effects to the diabetic perineuropathy and large vasculopathy.(4)The results of this article illustrate the TAP has a better protective effects than the simple recipe of the T and P.
