Expression And Significance Of Human Wings Apart-like And HPV In Cervical Cancer And Cervical Intraepithelial Neoplasia

Objective To investigate the expression and clinic significance of human wings apart-like and it's association with HPV infection in cervical cancer and cervical intraepithelial neoplasia.Methods RT-PCR and western blot were used to investigate the expression of human wings apart-like mRNA and protein in 75 cases with cervical intraepithelial neoplasia(including CC 18 cases,CINⅢ19 cases,CINⅡ18 cases,CINⅠ20 cases)and 16 normal cases.All cases didn't accept radiotherapy or chemotherapy.HPV was tested by HC2.Expression was evaluated by one-way analysis of variance and Student-Newman-Keuls analysis.Incidence was evaluated by chi-square test.There was statistically significant as P＜0.05.Results(1)The positive expression of hWAPL mRNA and protein was observed in different cases.The expression of hWAPL mRNA in CC,CINⅢ,CINⅡ,CINⅠ,normal tissues respectively was 1.21±0.76,1.16±0.49,0.52±0.31,0.30±0.12,0.21±0.08.The level of hWAPL protein in CC, CINⅢ,CINⅡ,CINⅠ,normal tissues respectively was 0.92±0.23,0.92±0.42,0.55±0.31,0.38±0.15,0.37±O.16.The level of hWAPL mRNA and protein were significantiy higher in CC,CINⅢtissues than in normal tissues(P＜0.01),and were higher in CC,CINⅢtissues than in CINⅡ(P＜0.01);There was statistically significant(P＜0.05)between CINⅡand normal tissues;There was no remarkably different between CINⅠ and normal tissues(P＞0.05),nor were they between CC and CINⅢ(P＞0.05).(2)The positive rate of HR-HPV in CC,CINⅢ,CINⅡ,CINⅠ,normal tissues respectively was 100%,100%,88.7%,30%,12.5%.The positive rate of HR-HPV in CC,CINⅢ,CINⅡwas remarkably higher than in CINⅠand normal tissues(P＜0.01);Among CC,CINⅢ,CINⅡtissues,There was no different either two compared with each other(P＞0.05).There was no different between CINⅠand normal tissues(P＞0.05);Conclusion The expression of hWAPh in cervical tissues is gradually increased with the lesion's severity.Upregulation of hWAPL may induce cervical cancer.There isn't different between CC and CINⅢ, which indicates that hWAPL is only related with precancerous lesion' s developing;There isn't different between CINⅠand normal tissues, which indicates that CINⅠisn't one passage of precancerous lesion' s developing.Only in special external factor such as HPV infection does it develop into cc.The positive rate of HR-HPV is higher in high grade squamous intraepithelial lesion(HSIL)tissues than in low grade squamous intraepithelial lesion(LSIL).Positive HPV results indicate potential danger of precancerous lesion,and is related to CC significantly.All show that hWAPL is hopeful to become a specific target for CIN diagnosis and screening and to elevate the accurate of screening combined with HPV.
