Isolation And Purification Of Compound Hawthorn Extract Using Versamide

In this dissertation, some influencing factors using versamide to isolate and purify compound hawthorn extract were investigated. Furthermore, technology scale up and optimize conditions of adsorption and desorption was obtained. It was provide experiment reference for realizing industrial production. The main results were focused on the following aspects:The testing methods of flavonoids and anthraquinone in compound hawthorn extract were established, and its technology was investigated.The wet method of packing column is decided by comparing dry method and wet method.In this paper, influencing factors of versamide adsorption and desorption extract was studied in caldron with agitation. The optimization conditions are crude drug concentration 0.255g.mL-1, pH 4, room temperature and adsorb time about 60 minutes. At the same time, elution of versamide was researched too, and the optimization conditions are 75% alcohol and eluting time 60 minutes. Versamide needn't be reactivated before repeating to be used 6 times.Influencing factors of versamide adsorption and desorption extract was study in fixed bed. adsorption rate 1BV/h, water eluting rate 3BV/h and alcohol eluting rate 1BV/h was decided. Moreover rules of scaling up is obtained, the relationship of adsorption quantity and ratio of high and diameter is qv= 4.1485-7.0625/(L/D). In these conditions, the yield of active components in compound hawthorn extract is exceed 90%, and dry weight remains 10% in extract. Finding unit adsorption quantity declines with column diameter amplifying in the conditions of same column high and adsorption rate.The technology of scaling up using versamide to isolate and purify compound hawthorn extract was established, and to decide equipment, material balance and productive flow sheet, and to direct company to establish flow sheet.
