| Objects:To study the effects of different factors on extraction and purification efficiency of hawthorn flavonoids by Macroporous Adsorption Resin, and obtain the most perfect method by single factor analysis and Orthogonal Test. And to study the effects of hawthorn flavonoids on the mouse alcohol liver.Methods:1. To study the effects of liquid solid ratio, extracting time, ultrasonic power and alcohol concentration on extraction technology of hawthorn flavonoids with single factor analysis, and to get the most perfect method by Orthogonal Test.2. To study the effects of flow rate, temperature, alcohol concentration and pH value on purification technonogy of Macroporous Adsorption Resin, and establish the most perfect method by Orthogonal Test.3. After the model of the mouse alcohol liver be formed, the change of aspartate transaminase (AST), alanine transaminase (ALT) in serum, superoxide dismutase (SOD), glutathione perioxidase (GSH-Px); and the content of malondialdehyde (MDA), nitrogen monoxide (NO), TNF-αbe investigated.Results:1. The perfect method of extraction for hawthorn flavonoids is A3B2C3D3, that is, using fifteen times of alcohol (85%) as solvent, extracting for 40min assisted by ultrasonic (700W).2. The perfect method of purification for hawthorn flavonoids is by D101 resin and A3B3C3D2, that is, using alcohol (75%)as eluent, eluting at the speed of 2.0mL/min with the condition of 80℃and pH(7.5).3. The effects of hawthorn flavonoids on mouse alcohol liver be investigated. The results showed that different doses of hawthorn flavonoids reduce the ALT and AST content of the mouse serum (P<0.05), and increase the GSH-Px and SOD content of the mouse liver tissue (P<0.05), and reduce the level of MDA, NO and TNF-α(P<0.05).Conclusions:1. The extraction method of hawthorn flavonoids in this study was better than the traditional ones. The average extraction rate is 4.736%. This method reduce the extraction time to a great degree, and reduce the extraction cost.2. The purification method of hawthorn flavonoids in this study was better than traditional. The purity of hawthorn flavonoids is 81.37%. The recovery rate of hawthorn flavonoids is 79.12%.3. Hawthorn flavonoids protect to some degree the mouse with alcohol liver by reducing the number of free radical in serum, and inhibit the lipid peroxidation. |
