Influence Of Breviscapine And Puerarin On The Expression Of Matrix Metalloproteinase-2 In Cultured Human Umbilical Vein Endothelial Cells

Objective: To investigate the effect of breviscapine and puerarin on the expression of matrix metalloproteinase-2 in cultured human umbilical vein endothelial cells. In order to provide theoretical basis for clinical application.Mehtods: (1).We detected the effect of breviscapine and puerarin on proliferation rate in cultured human umbilical vein endothelial cells (HUVECs) by MTT assay, then figured out the IC50 and IC10 respectively. (2).We selected three kind of concentrations of breviscapine(17.5ug/ml,35ug/ml,70ug/ml)and puerarin(250 ug/ml,500 ug/ml,1000ug/ml) to be coincubated respectively with HUVECs, and assess the effect of two kinds of drugs of different concentrations on the expression matrix metalloproteinase-2(MMP-2) in cultured HUVECs after 24 hours by immunocytochemistry. (3).The breviscapine of different concentrations(17.5ug/ml,35ug/ml,70ug/ml)were conincubated with cultured HUVECs for 24 hours after the HUVECs were stimulated in 1 hour by thrombin(4U/ml). The thrombin control group and normal control group were set as well. Measurements of MMP-2 was performed by immunocytochemistry.Results: (1).Compared with normal control group after 24 hours, group breviscapine(≤78.125 ug/ml) had no obvious effect to the proliferation of cultured HUVECs(P>0.05),but 156.25ug/ml,312.5ug/ml,625ug/ml,1250ug/ml breviscapin inhibited the proliferation of cultured HUVECs(P<0.01)with IC50=226.90ug/ml, IC10=73.99ug/ml;Also group puerarin (≤1250 ug/ml) didn't show any effect on the proliferation of cultured HUVECs(P>0.05),yet 2500ug/ml,5000ug/ml puerarin obviously inhibited it(P<0.01)with IC50=8350.94ug/ml, IC10=1117.37ug/ml. (2). Compared with normal control group, groups breviscapine had no obvious effect to the expression of MMP-2 in cultured HUVECs(P>0.05); groups puerarin increased the expression of MMP-2, There was difference between 250ug/ml puerarin and normal control group(P < 0..05),and there was significant difference between 500ug/ml,1000ug/ml puerarin and normal control group (P<0.01). (3). Compared with group thrombin , groups breviscapine could inhibit the expression of MMP-2 in cultured HUVECs which sitmulated by thrombin(P<0.01).There was no difference between group 35ug/ml and group70ug/ml (P>0.05).Conclusions: (1).Both group breviscapine(≥156.25ug/ml) and group puerarin (≥2500ug/ml) could inhibit the proliferation of cultured HUVECs. (2).Puerarin could increase the expression of MMP-2 in cultured HUVECs, but breviscapine couldn't. (3).Breviscapine could partially inhibit the expression of MMP-2 in cultured HUVECs stimulated by thrombin.
