Objective: This study was aimed to evaluate the expression of PTN,MK and CD34 and to find the differences of MVD in eutopic endometrium of endometriosis and normal endometrium of other disease,and to explore the etiopathogenisis of endometriosis.Methods: 40 cases of eutopic endometrium samples of endometriosis were studied,with 20 cases of proliferative phase endometrium,and 20 cases of secretory phase endometrium.Another 40 cases were collected of normal endometrium of other disease including 20 cases of proliferative phase endometrium and 20 cases of secretory phase endometrium as control.(1)The expressions of PTN,MK and CD34 were determined by immunohistochemical SP assay.(2)The microvessel density(MVD) in eutopic endometrium of endometriosis and normal endometrium of other disease was detected by quantitative determination.(3)PTN and MK expression in eutopic endometrium of endometriosis and normal endometrium of other disease was detected byWestern blot.(4) PTN and MKmRNA expression in eutopic endometrium of endometriosis and normal endometrium of other disease was detected by RT-PCR.Result:(1)PTN mainly expressed in the membrane and plasma of glandular epithelium of endometrium.PTN expression in study group was significantly higher than that in control group(P<0.01).PTN expression in secretory phase is higher than that in proliferative phase in 2 groups(P<0.05).MK mainly expressed in the plasma of grandular epithelium of endometrium and expressed in nuclei in some samples.MK expression in study group was significantly higher than that in control group (P<0.01).MK expression of proliferative phase in control group was higher than that of secretory phase (P<0.05), however, in study group,there was no significiant difference between proliferative phase and secretory phase(P>0.05).(2).The MVD of CD34 in eutopic endometrium of endometriosis was higher than that in control group(P<0.05).PTN was in positive correlation with MVD whether study group or control group.MK also was in positive correlation with MVD in 2 groups.(3) PTN and MK was dectected by Western-blot in 2 groups. Compared with control group, PTN expression in eutopic endometrium of endometriosis was significant higher (P<0.01); PTN expression in secretory phase is higher than that in proliferative phase in 2 groups (P<0.05).MK expression in eutopic endometrium of endometriosis was significant higher than that in control group(P<0.01), .MK expression of proliferative phase in control group was higher than that of secretory phase(P<0.05),however,in study group,there was no significiant difference between proliferative phase and secretory phase(P>0.05) (4) PTN and MKmRNA was dectected by RT-PCR in 2 groups . Compared with control group, PTNmRNA expression in eutopic endometrium of endometriosis was significant higher(P<0.01);.PTNmRNA expression in secretory phase is higher than that in proliferative phase in 2 groups(P<0.05).MKmRNA expression in eutopic endometrium of endometriosis was significant higher than that in control group(P<0.01), .MKmRNA expression of proliferative phase in control group was higher than that of secretory phase(P<0.05),however,in study group,there was no significiant difference between proliferative phase and secretory phase(P>0.05)Conclusions: PTN and MK expression in study group were higher than that in control group.The MVD was higher in eutopic endometrium of endometriosis than that in control group. Over-expression of PTN and MK in eutopic endometrium of endometriosis correlated to endometriotic cell angiogenesis.The changement of PTN and MK expression in eutopic endometrium of endometriosis may relate to the pathogenesis of endometriosis.
