Objecttive To study a new approach with 0% oxygen to establish the hypoxic-ischemic brain damage model of neonatal rat.,and to verify the superiority. Methods Wister rats of 7 days old were randomly allocated to 0% oxygen group(n=64),Rice-Vannucci group(n=24), ischemic group(n=24) and control group(n=24). The rats of 0% oxygen group were exposed to 0% oxygen at 37℃for 7 min. Unilateral Ligation of the left common carotid artery was carried out in the Rice-Vannucci group, then exposed to 8% oxygen at 37℃for 2 hrs. The ischemic group subjected to unilateral ligation of the common carotid artery without subsequent hypoxia.The control group subjected to neither ligation nor hypoxia.Every group was performed with assessment of brain water content, aprptosis of neurons and neurofunction.The brain was HE stained to observe pathological changs. Result The water content[(88.96±0.29)%] and aprptosis of neurons([31.52±5.45)%] of left brain in 0% oxygen group were significantly higher than those of ischemic group and control group(P<0.01),but there were no significant difference between 0% oxygen group and Rice-Vannucci group(P>0.05).The water content[(88.68±0.24)%]and aprptosis of neurons[ ( 30.85±5.38 ) %] of right brain in 0% oxygen group were significantly higher than those of Rice-Vannucci group, ischemic group and control group(P<0.01).Pathological changs was abnormal significantly in both side brain of 0% oxygen group.,but only was in left side brain of Rice-Vannucci group.No abnormalities were seen in ischemic group and control group.The neurofunction were impaired significantly both in 0% oxygen group and Rice-Vannucci group(P<0.01) Conclusion A simple and easy approach with 0% oxygen to establish the hypoxic-ischemic brain damage model in neonatal rat was found successfully.The new approach have more superiorities compared to other approach.
