Objective:To explore the influence of edaravone on caspase-3 and HIF-1 α expression in neonatal rats with hypoxic ischemic brain injury.Methods:Ten litters of newborn SD rats(with female rats)at 7 days of age were randomly selected and divided into 3 groups,a total of 108 rats.Thirty-six rats were randomly selected as control group(group A);the remaining 72 were subjected to hypoxic-ischemia treatment.Among the remaining 72 neonatal rats,36 rats were randomly selected as hypoxic-ischemic brain injury model group(group B).The remaining 36 neonatal rats were given edaravone intervention as the edaravone intervention group(group C).The three groups were divided into 6 subgroups according to the death time(6h,12 h,24h,48 h,72h and 5d),with 6 animals in each subgroup.Biological behaviors of all rats at different time points in each group were observed.The expression of Caspase-3 and HIF-1α and the apoptosis of brain cells were detected by HE staining,immunohistochemistry and TUNEL.Results:Compared with neonatal rats in group A at different time points,a number of neuronal apoptosis and the number of positive cells of caspase-3 and HIF-1α in cerebral cortex of neonatal rats in groups B and C were significantly increased at corresponding time points,the differences were statistically significant(P<0.05);Compared with neonatal rats in group B at different time points,the expression of apoptosis cells,caspase-3 and HIF-1α in cerebral cortex of group C were decreased,the differences were statistically significant(P<0.05).Conclusion:(1)Edaravone can relieve symptoms;(2)Edaravone can reduce the damage of cerebral cortex neurons;(3)Edaravone may play a neuroprotective role in reducing apoptosis by inhibiting HIF-1A and caspase-3 pathways.
