Purpose To measure the serum vascular endothelial growth factor(VEGF)and lactate dehydrogenase (LDH) levels in acute leukemia samples with different types and to explore the relationship between serum VEGF, LDH levels and curative effects in acute leukemia patients.Methods 45 cases of acute leukemia patients including 25 cases of primary leukemia patients, 20 cases of relapse and refractory patients, were enrolled in this study. Serum levels of VEGF and LDH were detected by enzyme-linked immunoabsorbent assay [ELISA] and automatic analyzer. The VEGF and LDH levels in the various subtypes of acute leukemia patients were contrasted to those of control groups. relationship between the VEGF , LDH levels and the numbers of the blast cells within nucleated cells In bone marrow and other related items were also analyzed. All data were analyzed by SPSS13.0. P Values less than 0.05 were considered to be statistically significant.Results (1)Serum VEGF levels of primary AL were (109.60±36.81)pg/mL, they were higher obviously than those(59.45±20.20)pg/mL of normal subjects and the differences were statistically significant(p<0.01),serum LDH levels of primary AL were (829.11±764.44)IU/L, they were higher obviously than those (141.07±40.11)IU/L of normal subjects and the differences were statistically significant (p<0.01). VEGF levels were reduced to(78.87±17.66)pg/mL in the AL patients with complete remission,no statistically significant differences to the control group(p>0.05), and LDH levels were reduced to(215.72±74.13)IU/L,no statistically significant differences to the control group(p>0.05). Serum VEGF levels of pretreatment patients in CR group were (101.09±32.44)pg/mL, and those of NR were(131.48±40.76)pg/mL, serum VEGF levels of NR group were higher than those of CR group. But the differences between them were not statistically significant( p>0.05). LDH levels of CR group were (735.88±713.52)IU/L, and those of NR group were (1068.86±895.48)IU/L, serum LDH levels of NR group were higher than those of CR group, But it was not statistically significant(p>0.05). (2)The VEGF levels of the relapse patients were (98.84±24.47)pg/mL and (133.48±60.96)pg/mL In complete remission phase and after relapse respectively, the differences were not statistically significant(p>0.05), The LDH levels were (284.60±74.36)IU/L and (602.20±200.70)IU/L In complete remission phase and after relapse respectively , the differences were statistically significant (p<0.01). (3)Positive correlation was shown between serum VEGF and LDH(r=0.651, p<0.01). The VEGF and LDH levels were related to the percentage of bone marrow blasts (r=0.521,p<0.01,r=0.519,p<0.01),but not related to the age, the white blood cell count (WBC) and hemoglobin concentration in the blood.Conclusions The serum VEGF and LDH levels of primary patients group were significantly higher than those in control group. It was reduced to the normal levels after complete remission, It rose again when relapsed. The VEGF levels in the refractory leukemia patients were much higher than those in the primary patients. The levels of VEGF and LDH were well corresponded with curative effects. So dynamically testing the change of serum levels of VEGF and LDH may help to get a good knowledge of the quantity of leukemia cell and the status of patients in certain degree. The levels of VEGF and LDH can be used as a guide to the assistance index of the response to treatment and the prognosis judgment of acute leukemia.
