Objective:To study the effect of hyperhomocysteinemia on cerebral ischemia reperfusion injury and to find the mechanisms happen during this process.Methods:78 healthy and male SD rats were randomly divided into 2 groups:Hhcy groups and the control group,39 rats was included in each.Plant feedstuff was provided to Hhcy groups and homomethionin feedstuff to the control group for 12 weeks.Measure the levels of the tHcy to be sure the Hhcy rat had been induced.Make up the models of cerebral ischemia reperfusion in rat with Hhcy by being subjected to 1,2,4 hours middle cerebral artery occlusion and 24 hours reperfusion,the groups that was subjected to 1,2,4 hours MCAO was named A group,B group and C group ordinally,13 rats was included in each group.Measure the neuroethology defect,the cerebral infarction volume,the levels of SOD,MDA and GSH-PX,patholoqical change of brain was observed with light microscope by HE stain and TUNEL stain.Result:1 The Hhcy group which was provided homomethionin feedstuff for 12 weeks was induced hyperhomocysteinemia.Mearsure the levels of tHcy at 0 week and the 12th week,at 0 week the Hhcy group(9.33±2.17μmol/L),the control group (9.15±2.29μmol/L),there is not statistically signigicant.(P>0.05).At the 12th week the Hhcy group(22.77±3.12μmol/L),the control group(9.10±2.26μmol/L),it is statistical significance;The levels of Hhcy group's tHey is higher at the 12th week than that at 0 week,there was no chang during 12 weeks about the control group.2 Analysis the result of the neuroethology defect,it showed the grades of Hhcy group about the neuroethology defect A group(10.44±0.72),B group(9.88±0.64),C group(9.30±0.95),they were lower than that of the control A group(11.56±1.23),B group(10.60±0.69),C group(10.33±0.71),it is statistical significance(P<0.05);The margin between the Hhcy group and the control group A group(1.11±0.13),B group (0.77±0.23),C group(1.03±0.26)is not statistically signigicant(P>0.05).3 The cerebral infarction volume of the Hhcy A group(43.41±5.29mm~3),B group (78.59±4.23mm~3),C group(125.54±8.48mm~3)is bigger than that of the control A group(33.86±7.33mm~3),B group(61.52±6.77mm~3),C group(91.60±4.7mm~3),it is statistical significance(P<0.05).The margin of the cerebral infarction volume between the Hhcy group and the control group A group(9.62±2.81mm~3),B group (17.07±2.65 mm~3),C group(33.53±4.71 mm~3)is statistical significance.4 The SOD levels of the Hhcy A group(276.03±55.44 U/ml),B group (224.33±31.95 U/ml),C group(176.34±39.12 U/ml)is lower than that of the control A group(338.72±29.06 U/ml),B group(314.75±47.52 U/ml),C group (290.48±17.32 U/ml),it is statistical significance(P<0.05).The margin of the SOD levels between the Hhcy group and the control A group(62.71±39.30 U/ml),B group (90.92±40.01 U/ml),C group(114.03±34.33 U/ml)isn't statistically signigicant.5 The MDA levels of the Hhcy A group(11.92±1.75nmol/ml),B group (17.45±6.22nmol/ml),C group(23.66±5.26nmol/ml)is higher than that of the control A group(5.71±2.75nmol/ml),B group(7.02±2.35nmol/ml),C group (10.21±3.23nmol/ml),it is statistical significance(P<0.05).The margin of the MDA levels between two groups A group(6.27±3.93nmol/ml),B group (9.09±4.00 nmol/ml),C group(11.40±3.43nmol/ml)is not statistically signigicant.6 The GSH-PX levels of the Hhcy A group(694.54±85.42)U,B group (417.94±74.82)U,C group(6.95±0.85nmol/ml)U,is lower than that of the control group,A group(1140.13±151.10)U,B group(933.70±296.15)U,C group (927.49±185.63)U,it is statistical significance(P<0.05).The margin of the GSH-PX levels between the two groups A group(445.59±193.23)U,B group (516.45±202.31)U,C group(516.69±123.25)U,is not statistically signigicant.7 The pathological change under the light microscope after HE stain about both the Hhcy group and the control group is the infarction in the Hhcy group is bigger than that of the control group.It shows that tissue looseness,karyopyknosis,disapper, cytopenia,the interspace of the cell and the blood vessel enlarges.8 Count the number of Apoptosis cell in unit visual field after TUNEL stain,the number of the Hhcy group is much than that of the control group.Conclusion:Hhcy increase the injury of the rat after cerebral ischemia reperfusion: The mechanisms may be accelerating oxidative stress,increase the production of the oxygen free radical and inducing cell apoptosis.
