Programmed Cell Death 5(PDCD5,formerly named TFAR19)is a new cell apoptosis related gene which was cloed from the human leukemia cell line TF-1 by the PEIKING UNIVERSITY GENG CENTER.PDCD5 is located at 19q12-q13.1,whole length of the gene is 6kb and includes 6 extrons and 5 introns,the cDNA is 590bp.The construction of deletion mutation and correlated function study of PDCD5 suggested that the extron 6 may be a function structure field that close related with promote apoptosis activity.Studies suggested that PDCD5 is a pro-apoptosis gene,the expression of PDCD5 protein is up-regulation and evident nuclear translocation can be watched in the morning of apoptosis,so it is possible to be a new marker of early apoptosis.The proto-oncogene B-cell lymphoma/Leukemia-2(BCL-2) was first cloned as the t(14,18)translocation breakpoint from human follicular center B-cell lymphoma by Tsujimoto in 1986.It is 230kb and includes 3 exons and 2 introns.This translocation moves the BCL-2 open reading frame from its normal promoter and regulatory sequences on chromosome 18 and places it under the control of the immunoglobulin heavy chain enhancer on chromosome 14,leading to the aberrant expression and over-expression of BCL-2 gene.BCL-2 can suppress apoptosis and prolong cell survival and it was the first reported anti-apoptosis gene.In order to investigate the significance of the expression of PDCD5 and BCL-2 protein and mRNA in multiple myeloma(MM),we analyzed 31 multiple myeloma and 25 cases of normal bone marrow as control by using immuneohistochemistry and Reverse transcription-poly merasechain reaction(RT-PCR).RESULT:By the immuneohistochemistry staining,the PDCD5 positive cell ratio(%)in every case of MM and control was 34.75±6.49and 52.98±5.84,respectively,SII(staining intensity index)was 281.16±75.33 and 462.84±39.77,the positive cell ratio and SII of PDCD5 is significant lower in MM than that in control group(p<0.05).The BCL-2-positive cell ratio(%)in every case of MM and control is 29.97±5.57 and 5.56±1.95,SII is 224.94±57.72 and 27.84±9.75,respectively.The positive cell ratio and SII in MM is significant higher than in control.The expression of PDCD5 and BCL-2 protein is negative-related(r=-0.86,P<0.05).The specific value of PDCD5 mRNA andβ-actin mRNA is 0.33±0.07 and 0.53±0.05 in the two groups respectively,the relative level of PDCD5 mRNA in MM is significant lower than in control(p<0.05),the ratio of BCL-2 mRNA andβ-actin mRNA is 0.33±0.08 and 0.12±0.02,the relative express level of BCL-2 mRNA is significant higher than that in control(p<0.05).The level of PDCD5 and BCL-2 mRNA is negative-related(r=-0.90,P<0.05)CONCLUSION:In multiple myeloma,the expression of protein and mRNA of PDCD5 is down-regulated and BCL-2 up-regulated,the expression of the two gene is negative-related.
