Objective : By animal experiment to research the effect of the knee joint medial collateral ligament (MCL) reconstruction using the tendon autograft, which has influenced the degradation of the cartilage of knee joint. The purpose of this research is to provide experimental evidence on healing serious injury of MCL using the reconstruction of tendon in clinic, in addition to reduce the osteoarthritis.Methods: take twenty-nine ripe New Zealand rabbit. Cut knee joint medial collateral ligament (MCL) of both sides from the middle, in order to create contrast self models. Select random experimental group. Use simulating semi-tendinous tendon on the reconstruction of MCL to do surgery. The contrast group did not carry out the reconstruction surgery. Six rabbits were enthanised separately in 4, 12, 16,24 weeks after surgery. Through histological section, immunity histological chemistry and electron microscope, observe the languishment of the cell of knee joint cartilage and the expression of languishment control gene BCL-2.Results: 1.Histological observation: the external situation and arrangement of cartilage of 4w, 12w, 16w, 24w are better than contrast group. There are lots of chondrocytes which are vanished in 24w contrast group. 2. Immunity histological chemistry: there are BCL-2 masculine chondrocytes in both experimental group and contrast group. Compared with contrast group in the experiment, the percentage of BCL-2 masculine chondrocytes is distinct (P<0.05). 3. Transmission electron microscope. Every experimental group only find some degenerative chondrocytes. All contrast groups have both languished and apoptosis is especially serious in 24w group.Conclusion: 1.After severe injury of MCL, use the techniques of reconstruction of semi-tendinous to maintain the arrangement and configuration of chondrocytes. 2.Reconstruct MCL by semi-tendinous after severe aspiration injury, which could slow down the languishment of cartilage, and decrease apoptosis of chondrocytes.
