Objective:To investigate whether pregnancy-associated plasma protein-A had effects on the injury of vascular endothelial cells(VEC) and whether Statins could protect vascular endothelial cells from the injury that made by the Pregnancy-associated Plasma Protein-A.Method:HUVECS cell line,derived from human umbilicalvein endothelial cells,were cultured in vitro with pregnancy-associated plasma protein-A with concentrations of 0 ng/ml,25 ng/ml,50 ng/ml,100 ng/ml for 0,12,24,48 hours,respectively.Then 10-7mmol/L,10-6mmol/L, 10-5mmol/L fluvastatin were added first into vascular endothelial cells (VEC)for 20minutes.And secondly vascular endothelial cells(VEC) were cultured with pregnancy-associated plasma protein-A with concentrations of 200 ng/ml for 12 hours.At the same time the mean optical density(MOD)at each concentration was determined in order to calculate the influence.NO levels and ET levels were determined by using spectrophotometer and immunehistory.Results:(1)In PAPP-A groups,the NO levels of HUVEC cells were significantly lower than those of the control group(P<0.05).At the same time the ET-1 levels of HUVEC cells were significantly higher than those of the control group(P<0.05).The changes were all dose-dependant.But at the same concentrate there was no significant difference with time.The immunehistory effects of ET-1 were that in PAPP-A groups,we could see positive result,but in the control group,we could only see negative result.(2)ln fluvastatin groups with PAPP-A,there were significantly change in them to the PAPP-A group,the NO levels of HUVEC cells were significantly higher than those of the PAPP-A group(P<0.05).At the same time the ET-1 levels of HUVEC cells were significantly lower than those of the PAPP-A group(P<0.05).The changes were all dose-dependant.Conclusions:1.Pregnancy - associated plasma protein-A had effects on the injury of vascularendothelial cells.The changes were all dose-dependant and had no significant difference with time.2.Fluvastatin could protect vascular endothelial cells from the injury that made by the pregnancy-associated plasma protein-A.The effects were all dose-dependant.
