| Objectives To investigate the effect of fluvastatin on endothelin-1 (ET-1) and endothelin A receptor (ETaR) in rat after acute myocardial infarction (AMI),whether fluvastatin can decrease myocardial infarct size.Methods 1.Experiment objective:50 male SD rats weighted 230 gram to 270 gram.2. Experiment group:All animals were randomly divided into 3 group,named N,C,F. Rats of N were nature control group, which for sham-operated were only opened thoracis (n=10). Rats of C for control group in which were divided into Cl(n=10) and C2(n=10).Rats of F for fluvastatin-treated group in which were divided into Fl(n=10) and F2(n=10).Cl and Fl were established for detecting myocardial infarct size. C2 and F2 were established for detecting biochemistry parameters. The group F (fluvastatin group) was administered fluvastatin suspension (20 mg/kg/day) by oral gavage for 4 days .Rats of N and C were administered normal saline(2ml/day).After opening thoracis and pericardium in rats of F and C, the left anterior descending coronary artery (LAD) was ligated near its origin using a 7-0 silk suturen after 4 days,, rats of N had a similar surgical procedure except that the suture was not tightened around LAD.3.Detective parameter:â‘ ECG was measured after ligated LAD 5min.The blood was drawn after ligated LAD 230min. Plasmic CK,CK-MB,ET-1 was measured from it.
