Chapter one:The role of L-a-aminoadipic acid played in the changes of the GFAP expressed in retinal Miiiler cells.Objectives:To investigate the changes of the Glial fibrillary acidic protein(GFAP)expressed in retinal Müller cells are affected by L-a-aminoadipic acid(L-α-AAA).Methods:20 guinea pigs were divided into two groups,that is,group A(experimental)and group B(control),there were 10 guinea pigs in each group.In group A,L-α-AAA would be injected into vitreous of right eyes. In group B,saline would be injected into vitreous of right eyes.Test changes of the GFAP expressed in retinal Müller cells by Immunohisto-chemistry.Result:Semi-quantitative analysis of immunohistochemistry slice showed that the positive value of gray area in group A(145.28±10.06)and group B(163.67±11.39)compared to a statistically significant difference(P<0.05).Conclusion:L-α-AAA which result in GFAP up-regulation expressed in retinal Müller cells can induce retinal Müller cells damage effectly. Chapter two:The role of L-a-aminoadipic acid played in the changes of the refractions in FDM of guinea pig's eye.Objectives:To investigate the changes of the refractions in Form deprivation myopia(FDM)of guinea pig's eyes are affected by L-a-aminoadipic acid(L-α-AAA).Methods:FDM in guinea pigs was induced by covered right eye with translucence eye shield.50 guinea pigs were divided into 5 groups,that is, group A(normal),group B(2wks eye-covered),group C(2wks eye-covered+1wks eye shield removed),group D(2wks eye-covered+ L-α-AAA),group E(2wks eye-covered+PBS).There were 10 guinea pigs in each group.Except group A,its contralateral uncovered eye served as self-control ones.Ocular diopters and axis oculis of eyes were determined by retinoscopy after enlargeing pupil and A-scan ultrasonography.Test changes of the GFAP expressed in retinal Muller's cells of each group by Immunohisto-chemistry.Result:1.Changes of Ocular diopters and axis oculis in FDM eyes.Ocular diopters and axis oculis of all covered eyes in groupB,C,D,E and normal eyes compared to a statistically significant difference(P<0.05), groupB,C,D,E and its self-control eyes compared to a statistically significant difference(P<0.05).Group D and group B compared to a statistically significant difference(P<0.05).Ocular diopters of all covered eyes in group C of 2wks and group C of 3wks compared to a statistically significant difference(P<0.05).2.Expression of GFAP in the MOiler cells of retinaSemi-quantitative analysis of immunohistochemistry slice showed that the positive value of gray area in group B,C,D,E(129.34±11.23, 145.25±13.33,103.35±15.54,133.12±12.95)and groupA(161.55±12.05), compared to a statistically significant difference(P<0.05).Group D (103.35±15.54)and group B(129.34±11.23)compared to a statistically significant difference(P<0.05).Conclusion:1.After 2wks eyes be covered can result in FDM of guinea pig's eyes accompaned by ocular diopters and axis oculis increasing,de-covered eyes lwks accompaned by ocular diopters decreasing.2.Covered eyes into which L-α-AAA been injected accompaned by ocular diopters and axis oculis decreasing.
