Objective:To observe the expression of angiopoietin-1(Ang-1), angiopoietin-2 (Ang-2)and their acceptor Tie-2 proteins in Juvenile nasopharyngeal angiofibroma(JNA).Meanwhile, to study the relationship among Ang-1,Ang-2 and tumor's microvessel density, and to study their influence on the tumor's vascularization and dilatancy growth.Method: Ang-1, Ang-2 and their acceptor Tie-2 proteins were measured by immunohistochemistry stain from 30 JNAs and 20 chronic nasopharyngeal inflammatory tissues for control. Vascular endothelial cell marked by CD34 to calculate the microvessel density of JNA and chronic nasopharyngeal inflammatory tissue. To analyze the dependability among Ang-1﹑Ang-2 and MVD in the peripheral tissue of 20 JNAsResult:Ang-1﹑Ang-2 and Tie-2 proteins were mainly expressed in cytoplasm.The positive rate of Ang-2 and Tie-2 proteins was 76.67% and 100% in vascular endothelial cell and fibroblast of JNA respectively. Obviously higher than that in chronic nasopharyngeal inflammatory tissue.But the Ang-1 positive rates were all 80% in vascular endothelial cell , fibroblast of JNA and chronic nasopharyngeal inflammatory tissue. According these results,there was no statistically significant between them. The microvessel density in JNA was lower than that in chronic nasopharyngeal inflammatory tissue.(P<0.05).The expression rate of Ang-2 protein had some relationship with clinic staging of JNA(P<0.05),and the higher clinical staging of JNA, the higher expression rate of Ang-2 protein.In the periphery tissue of 20 JNAs, a negative correlation between Ang-1 protein and Ang-2 protein was observed. (r=- 0.4654; P <0.05), while the relation between Ang-2 protein and MVD was positive (r= 0.5083, P < 0.05).NO relation between Ang-1 protein and MVD(r=- 0.3241, P >0.05).Conclusion: Ang-2 protein is over-expression in JNA and the Ang-2 has positive correlation with MVD in the periphery tissue of JNA , the results suggest that Ang-2 promotes angiogenesis and tumor's dilatancy growth in JNA. Ang-1 protein has no relationship with angiogenesis and tumor's dilatancy growth in JNA. The over-expression of Tie-2 protein may has some relationship with JNA'S angiogenesis. We suppose that the role of Ang-2 is dominant in the effect of Angs and their receptor on angiogenesis and tumor's dilatancy growth in JNA.
