| Objective: To investigate the effect of blockage of renin-angiotensin systemfor Angiopoietin-1、Angiopoietin-2、Tie-2expression on renal and serum indiabetes nephropathy rats.Methods:Sixty male SD rats were took part in this trial. Random drawed10ofthem as normal control(group A),fed with normal food. The last fifty fed withhigh fat and sucrose diets.After4weeks,the higt fat and sucrose rats injected withlow dose streptozotocin（35mg/kg）to establish diabetes nephropathy model. Aftersuccess established the DN animal model, the experimental rats were randomlydivided into four groups, diabetic nephropathy model group(group B),treat withbenazepril group(group C),treat with irbesartan group (group D),treat withbenazepril and irbesartan group (group E). each group of ten rats. After8weeks，all rats been executed. Kindney weight/body weight, urea nitrogen，serumcreatinine，24-hours urine protein, and were measured to evaluate renal function.Angiopoietin-1、Angiopoietin-2、Tie-2in the intrarenal was stained by immunohistochemical(IHC)technique, which to detected theirs content in serumby ELISA.Conclusions: Rats of one model group after having the high glucose and high fatdiet，were intraperitoneally given low dose streptozotocin，can successfulinducted to type2diabetes mellitus model. Combination of benazepril andirbesartan offer futher benefits arrest proteinuria and protect from renal functionand structure impairment. It is suggested that the expression of Angiopoietin-1、Angiopoietin-2、Tie-2in serum and renal tissues of rats with diabeticnephropathy is increased than normal contral. And blackage of RAS in DN ratswith benazepril and irbesartan could reduce the expression of Angiopoietin-1、Angiopoietin-2、Tie-2in serum and renal tissues.Especially, combination ofbenazepril and irbesartan treatment had a extremely effect on reduce theexpression of intrarenal Angiopoietin-1、Angiopoietin-2、Tie-2. |
