The fabrication of the Guided Tissue Regeneration(GTR)membrane materials have become the key technique of the tissue engineering scaffold study.The cells adhere well on the fibers whose dimension is below their own (microscale)so that the porous three dimension electrospun scaffold material can mimic the structure of the natural extracellular matrix better and have the potential to be an ideal GTR membrane material.The electrospun natural polymer fiber membrane,which is porous,porosity adaptable and biocompatible,has great application in the GTR membrane material field.Collagen is the major protein component of bone,cartilage,skin and connective tissue and also the major constituent of all extracellular matrices in animals.It has many fine physics,the chemical properties,like the sol and the gelatin reversible transformation,side chain groups activeness is high,the amphoteric electrolyte characteristic,compares with the collagen,cost more inexpensive and so on.Recently,uses of gelatin-based biomaterials in applications of blood vessel repair,the medicine transmission carrier,the flesh wound to spread governs and so on have been reported.Gelatin has the water affinity,so it must be able to use after crossing linking.EDC/NHS is one kind of water-soluble crosslinking agent.Because the crossing linking intermediary product and itself may dissolve in the water which will not introduce the toxic ingredient in the crossing linking matrix,it was used as one kind of ideal mix biology crosslinking agent.But as a result of the gelatin water affinity,the water introduction in the crossing linking environment can create the gelatin nanofiber mat fierce contraction,the curl,and affects the cross-linkage reaction advance which will not easily get crosslinked gelatin nanofiber mat flatly,smoothly and high degree of cross linking.In this paper,we crosslinked gelatin nanofiber mats in different rate of alcohol/water solutions with the same concentration EDC/NHS.According to the analysis on their macroscopic performance,microcosmic performance, swelling,dissolution and mechanical properties,the research demonstrated that gelatin nanofiber mat which crosslinked in the absolute alcohol solution containing the best overall performance.We also made research on its behavior in vitro degradation process,and finally found the gelatin nanofiber membrane may obtain the widespread application in the biological medical material domain after this kind of crossilinking.The research demonstrated that the gelatin solution's viscosity reduces greatly and is easy to obtain continuously,thick thin even gelatin nanofiber under the temperature of 44℃. EDC/NHS crossinglinking gelatin nanofiber mat optimized condition:In 20℃, the concentration of EDC/NHS is 1.5wt%,MEDC/MNHS=5/2,alcohol and the water volume ratio is 99.7:0.3,the crossing linking time is 48h.After the crosslinking,the gelatin nanofiber mat surface is flat and smooth,the crosslinking degree is high,the tensile property enhances obviously,the swelling and dissolving performance are good.Under the research on the degeneration performance of gelatin nanofiber mat which crosslinked in optimization crosslinking condition,we found that biodegradation performance of this membrane material is good.It could degrades completely after 40 days,and the change of its pH value,swelling and dissolving properties are not big that could reduce the aseptic inflammation and is advantageous to its application as a kind of biological medical material.
