| ObjectiveBy using PC12 cells as the model of dopaminergic neuron, this article observed the cell growth, proliferation and apoptosis of PC12 cells,detected the expression and location of Hsp70,Survivin,Bcl-2 and Bax proteins in PC12 cells, and explored the possible mol-mechanism of Manganese-induced apoptosis in PC12 cells.MethodsThrough establishing PC12 cells as the model treated by manganese chloride,the morphological changes of PC12 cells induced by Manganese chloride were observed with microscope; MTT colorimetry tests were performed to exam cell growth and livingness of PC12 cells in various dose of Manganese; apoptosis of PC12 cells was detected by using flow cytometry (FCM) and agarose gel electrophoresis(AGE) ; immunocyto chemistry was used to measure the expression and location of proteins of Hsp70,Survivin,Bcl-2 and Bax.ResultsAfter treatment with Manganese chloride for 24 hours,PC12 cells showed evident morphological changes; Manganese might suppress the proliferation of PC12 cells in dose and time-dependent manner and induce PC12 cells apoptosis in dose-dependent manner; treated with Manganese chloride (400 umol/L) for 48 hours, the proliferation- inhibition ratio of PC12 cells was 58.4%. Manganese boosted the expression of Bax in dose-dependent manners, which had a positive correlation with PC12 cells apoptosis(0.972,P<0.01); while Manganese dose- dependently inhibited the expression of Hsp70,Survivin and Bcl-2 in PC12 cells , which had a negative correlation with PC12 cells apoptosis(R2=-0.990, R3=-0.976,R4=-0.980,P <0.01). Conclusions Acting on PC12 cells for a while, Manganese could inhibite the growth and livingness of PC12 cells in dose and time-dependent; PC12 cells were induced to apoptosis when they were exposed to Manganese chloride for 24 hours, and had dose-dependent manner; Manganese boosted the expression of Bax proteins which had a positive collelation with apoptosis ratio , and inhibited the expression of Hsp70,Survivin and Bcl-2 proteins which had a negative correlation with apoptosis ratio; Manganese could induce PC12 cells apoptosis,which might be caused by several factors, and the apoptosis was induced by the enhanced expressions of Bax and the prohibited expressions of Hsp70,Survivin,Bcl-2 in PC12 cells. |
