ObjectiveTo study the anti-tumor effect and explore its mechanisms of arsenic trioxide(As2O3) on the human Gastric cancer cell line SGC-7901. It was also observed the expression of vascular endothelial growth factor (VEGF) and Nuclear factor kappa B (NF-κB).MethodsThe human gastric cancer cell line SGC- 7901 was treated in vitro using As2O3 ,and then the effect of cell growth inhibition was assayed by MTT method.Cell cycles were analyzed by flow cytometry(FCM)and the expression of NF-κB, VEGF was deteced by immunocytochemistry .Results(1) As2O3 could inhibit the growth of SGC-7901 and the inhibitory effect was dose-dependent minner (P<0.05)(2) Flow cytometry analysis revealed that compared with control group ,there were no significant difference in experimental group which cells were treated with 1μmol/LAs2O3 alone for 48 hours in apoptosis (P<0.05).Compared with control group, after 48 hours treatment of 2.5,5μmol/LAs2O3 , there was significant apoptosis on SGC-7901 cells (P<0.05).There was a dose-dependent manner.(3) The cell cycle distribution was altered and the ratio of GO/G1 phase cell decreased. The percentage of cells in G2/M phase increased significantly(P<0.05)(4) Compared with control group, NF-κB and VEGF expression was downregulated.There was correlation between the expression of NF-κB and VEGF (r=0.864, P<0.05)Conclusion(1) As2O3 has the synergistic effects of the anti-gastric cancer and which may be caused by arresting cell cycle and inducing apoptosis. The apoptosis effect was dose-dependent minner.(2) NF-κB expression was downregulated and which may be the mechanisms of As2O3 inducing apoptosis.(3) As2O3 could down-regulation expression of VEGF.There was correlation between the expression of NF-κB and VEGF.
