The Effect WIN55, 212-2 To Intraocular Pressure(IOP) In Rabbits

Background:Glaucoma is one of cause of blindness clinically with the damage of optic papillary and retinal never fiber layers and glaucomatous visual field loss.At present the treatment method of glaucoma is still reducing IOP.Cannabinoids have been highly paid attention because of their unique reducing IOP and neuroprotective properties,but its systemic side effect is large,and it is lipid solubility,its solubility is very low in aqueous.so its reducing IOP effect is not obvious after local medication,recently people discovery that tocrisolve could increased its solubility.Objective:Cannabinoid was dissolved into tocrisolve and topical ophthalmic use,respectively observe reducing intraocular pressure effect to normal and glaucoma domestic rabbits after local medication and evaluate its side effect.Methods:1,Ten normal rabbits were randomly divided into two groups,one group right eye was received WIN55,212-2,another group was received TocrisolveTM 100 and left eye was received saline as contral.Drawing IOP variation curves following time after medication within 6 hours.Then right eye was received WIN55,212-2,left eye was received saline at 8Am,2Pm,8Pm,IOP of both eyes was measured at 8Am,10Am,4Pm,8Pm everyday within 3 weeks,and observation the variation of anterior ocular and fundus after continuous medication.2,Twenty-four rabbits right eyes with carbomer-induced elevation of IOP were randomly divided into 3 groups.Left eye was a blank control group.Each right eye conjunctival sac in every group received topical application of 0.125%WIN55,212-2 ophthalmic formulations, 0.9%NS,and 0.5%timolol.IOP was measured once everyday after topical ocular administration of the ophthalmic formulations.Results:1,IOP was significantly decreased half an hours after the application of WIN55,212-2in normal rabbit eyes(p＜0.01),and appeared the peak value of the reducing IOP after receiving the eye drop for 0.5-1.5hours and back to baseline level after 5 hours.And TocrisolveTM 100 have not effect of the reducing IOP,have no significantly difference compared base IOP.The decreased was maintained during 3 weeks treatment period,there was no obviously abnormality in anterior ocular and fundus after continuous medication.2,IOP decreased rapidly by up to 45%in 0.125%WIN55,212-2 eye drop treatment group ＞60%in 0.5%timolol treatment group after 3weeks,The decreased was maintained during the treatment period.The maximum reduction in IOP was significantly different among the groups.Conclusions:The IOP lowing effect of 0.125%WIN55,212-2 is markly in nomal and glaucomatous rabbits eyes.After the application of 0.125%WIN55,212-2,the decreased was maintained during the treatment period.And there was no significantly side effects.
