A successful pregnancy reflects actually materno-fetal immune tolerance. The mother does not attack the fetus that carries paternal antigen until its labor. This process depends a unique materno-fetal immune milieu, especially growth and invasion of trophoblast; otherwise, miscarriage is inevitable.KAI1/CD82 is a metastasis-suppressor gene expressed in a lot of tumors, and plays an important role in suppressing the viability, invasiveness and metastasis of the cancer cell. The trophoblasts at the materno-fetal interface possess the ability to invade into the decidual tissue, but compared with the tumor cell, the invasiveness of the tropholbasts must be strictly restricted. To investigate the regulation of KAI1/CD82 in the materno-fetal interface, the present study explored the expression and role of KAI1/CD82 at the materno-fetal interface in the first trimester pregnancy.Partâ… The expression of the metastasis-suppressor gene KAI1/CD82 at the materno-fetal interfaceObjective: To evaluate the expression of the metastasis-suppressor gene KAI1/CD82 in the decidua and the villus and the association with RSA.Methods: RT-PCR and Real-time PCR were used to study the transcription of KAI1/CD82, and Western-blot and immunohistochemistry to explore the translation of KAI1/CD82 in the decidua and the villus from the RSA with the normal early pregnancy as control.Result: The mRNA level of KAI1/CD82 in the villus and the decidua of RSA was significantly lower than that of the normal early pregnancy, while the protein level of KAI1/CD82 in the decidua of the RSA is significantly lower than that of the normal early pregnancy. The protein translation of KAI1/CD82 was not found in the villus.Conclusion: The metastasis-suppressor gene KAI1/CD82 in the decidua regulates the invasion of the trophoblast, and over-expression in the decidua would lead to RSA. Partâ…¡The decidual stromal cells regulate the invasion of the trophoblast via expressing KAI1/CD82Objective: To explore the regulation of KAI1/CD82 in the cross-talk between decidual stromal cells and the trophoblasts.Methods: The co-culture system including the endometrial stromal cells and the BEWO cell lines was established to investigate regulation of the decidual stromal on the invasiveness of trophoblast cells. RNAi was used to knock down KAI1/CD82 in the ESCs to investigate effects of the ESC-expressed KAI1/CD82 on the invasiveness of trophoblast cells.Result: The silencing of KAI1/CD82 was constructed in ESCs. The invasiveness of the BEWO trophoblast cell line was increased significantly after the KAI1/CD82 on ESCs had been silenced in the coculture.Conclusion: The KAI1/CD82 expressed in the decidual stromal cells could suppress the invasion of trophoblast cells.Partâ…¢The KAI1/CD82 was not involved in the CsA-driven invasiveness improvement of trophoblastObjective: To explore if KAI1/CD82 was involved in effects of CsA on trophoblast invasiveness.Method: After the decidual stromal cells were isolated, FCM and the In-cell Western were used to analyze the expression of KAI1/CD82 on the decidual stromal cell after the treatment with CsA.Result: The expression of KAI1/CD82 on the decidual stromal cell after the treatment with CsA did not show any change.Conclusion: the CsA does not influence the expression of KAI1/CD82 on the decidual stromal cell.
