Research objectiveOrthogonal experiment elicited a rational combination plan of swimming exercise,vitamin C and vitamin E to prolong the lifespan of S180 sarcoma mice, namely, VC 500mg/kg +VE 50mg/kg+1/2exhaustion swimming time, basing on his study, this paper researched mainly the inhibitory effects in vivo of this rational prescription on S180 cells, assay the effects of this prescription on free radical indexes, SOD,GSH-PX activities ,and MDA content in serum,liver tissue, and tumor tissue of tumor mice, observe its effect on inducing apoptosis of tumor cell, validated its creditability of the result of prolonging lifespan. Aims to provide reference about the cooperation actions between vitamin C+E and swimming exercise, to prolong lifespan and improve survival quality of tumor.Research methodsBy means of transplantable tumor ,each mice was injected 200 thousand units,0.2ml tumor dilution liquid on its right outside armpit, objects of the experimentation were 64 female Balb/C mice,13weeks old , 20±2g, after inoculation S180 sarcoma cells, mice were divided into 4 groups randomly,with 16 mice in each group(8 to be cut ,8 to be left to observe lifespan),they were contrast group(C),swimming + vitamin C+E group(TVCE),pure swimming group(T), and pure vitamin C+E group(VCE),On the fifth day , All the mice take a swim about 30 minutes long ,but on the next two days ,only mice in groupsⅡandⅢtake adaptable swimming exercise ,since the eighth day, each group is to carry on the experiment plan; on 43rd day ,8 mice each group were executed,and the left were going on the experiment till the 49th day, to observe lifespan. Detect free radical indexes with free radical reagent boxes, flow cytometry was used to detect the apoptosis index and proliferation index in tumor tissue of this best prescription. The data are analyzed by SPSS11.0,and showed with average±sd, with the methods of univariate analysis, the diversity is 0.05.Research results⑴the antineoplastic rate of swimming + vitamin C+E group,pure swimming group, pure vitamin C+E group were 68.66%,58.08%,21.06%,respectively.⑵tumor weights, lifespan in each group was: contrast group, 10.21±3.53g, 40.69±6.18d; swimming+ vitamin C+E group,3.20±1.20g, 50.19±11.25d;pure swimming group, 4.28±1.84g, 41.77±5.64d; pure vitamin C+E group, 8.06±1.90g, 43.08±5.38d.⑶Free radicals in serum: compared with contrast group, activities of SOD and GSH-PX in pure vitamin C+E group and swimming + vitamin C+E group had significantly increased (P﹤0.01);compared with contrast group, pure vitamin C+E group, MDA content in swimming + vitamin C+E group had decreased(P﹤0.05),while compared with pure swimming group, MDA content in swimming + vitamin C+E group had significantly decreased(P﹤0.01);compared with pure swimming group and pure vitamin C+E group, SOD activity in swimming + vitamin C+E group had significantly increased (P﹤0.01), and MDA content in swimming + vitamin C+E group had decreased, their differences were (P﹤0.01)and(P﹤0.05);compared with pure swimming, GSH-PX activity in swimming + vitamin C+E group had significantly decreased(P﹤0.01).⑷Free radical in liver tissue: compared with contrast group, GSH-PX activity in swimming + vitamin C+E group, pure swimming group had significantly increased(P﹤0.01);compared with contrast group, MDA content in swimming + vitamin C+E group ,and pure vitamin C+E group had decreased, their differences were(P﹤0.05)and (P﹤0.01); compared with pure swimming group and pure vitamin C+E group, GSH-PX activity in swimming + vitamin C+E group had increased , their differences were(P﹤0.05)and (P﹤0.01); compared with contrast group ,SOD activity in the three experimental groups had not significantly increased(P﹥0.05).⑸Free radical in tumor tissue: compared with contrast group, GSH-PX activity in swimming + vitamin C+E group, pure swimming group had significantly increased(P﹤0.01); compared with contrast group, SOD activity in swimming + vitamin C+E group,and pure vitamin C+E group had significantly increased(P﹤0.01), MDA content had significantly decreased(P﹤0.01),and SOD activity, MDA content in pure swimming group had not significantly changed; compared with pure swimming group, SOD activity in swimming + vitamin C+E group had significantly increased(P﹤0.01), MDA content had decreased(P﹤0.05); compared with pure vitamin C+E group, GSH-PX activity in swimming + vitamin C+E group had significantly increased(P﹤0.01).⑹compared with contrast group, apoptosis index in three experimental groups had decreased ,but without significant difference(P﹥0.05),compared with pure vitamin C+E group, swimming + vitamin C+E group had a relatively higher apoptosis index(P﹤0.05);compared with contrast group, proliferation index in pure swimming group had decreased(P﹤0.05); compared with pure swimming group, proliferation index in swimming + vitamin C+E group had increased(P﹤0.01).Conclusions⑴This rational prescription of swimming + vitamin C+E has a positive effect to prolong lifespan of the tumor mice, and to reduce tumor weight, body weight.⑵This rational prescription of swimming + vitamin C+E would enhance the ability of antioxidation, and reduce MDA content in serum and liver so as to protect tumor economy against free radical attack.⑶This rational prescription of swimming + vitamin C+E has an effect in inducing apoptosis to some extent, it might be one of the causes to explain its prolonging lifespan in reason.
