【Purpose】1.To approach the role of NF-κB in acute pancreatitis associated lung injury.2.To investigate the influence of resveratrol on the action of NF-κB in acute pancreatitis associated lung injury,and investigate the effects and mechanism of Resveratrol in severe acute pancreatitis(SAP)and severe acute pancreatitis(SAP) associated acute lung injury(ALI).【Method】1 the subgroup of the animals A total of thirty Sprague-Dawley ratswere randomly divided into sham operation group,SAP group and Resveratrol treatment group.2 the preparation of the model and the index to be detected The model of SAP was established by retrograde pancreatic injection of sodium taurocholate(1ml/kg). Opening the abdomen and massaging the pancreas lightly in SO group.The resveratrol sodium(10g/l)was given through vena caudalis after modeling(0.1ml/100 g).DMSO was given through vena caudalis at the same time after modeling(0.1ml/100 g).The rats were killed at 6 hours after modeling;the ascites volume,contents of amylase,arterial oxygen saturation and wet/dry were determined,The activation level of NF-κB of pancreas and lung was detected by immunohistochemistry with SABC.【Results】1.Ascites volume,AMS level,pancreatic and pulmonary wet/dry weight ratio in each group:The ascites volume,AMS level,pancreatic and pulmonary wet/dry weight ratio of pancreatic group was remarkably higher than the sham operation group(P<0.01),the ascites volume,AMS level,pancreatic and pulmonary wet/dry weight ratio of resveratrol treatment group was higher than the sham operation group,but it was much lower than SAP group.(P<0.05).The arterial oxygen saturation in SAP group was lower than SO group(P<0.01),the arterial oxygen saturation in RES group was lower than SO group,but it was much higher than SAP group(P<0.05).2.The.histological grading of pulmonary and pancreatic in each group:No obvious pathological changes were found in pancreatic and pulmonary tissue of sham operation group under microscope,but it could be found in SAP group.There weren't obvious pathological changes in pulmonary tissue of resveratrol treatment group,pathological injuries of pancreas in resveratrol treatment group were significantly less than in SAP group(P<0.05).3.The expressions of NF-κB in each group:The expressions of NF-κB in pancreatic and pulmonary tissues could hardly be found in sham operation group.In SAP group and resveratrol treatment group,the expressions of NF-κB in pancreatic and pulmonary tissues remarkably increased compared to those in sham operation group,and they decreased much more in resveratrol treatment group than that in SAP group(P<0.05).4.The analysis about dependability:The NF-κB action of pancreas is positive correlation to the histological grading and wet/dry weight ratio of pancreatic,r=0. 632,r=0.687(P<0.01);The NF-κB action of pulmonary is positive correlation to the histological grading and wet/dry weight ratio of pulmonary,r=0.587,r=0.635(P<0.01).【Conclusion】NF-κB was obvious activated in pancreatic and pulmonary tissues,it play an important role in acute pancreatitis associated lung injury;Resveratrol plays a role in the treatment of SAP and SAP associated ALI.It may depress the expression of NF-κB and suppress the inflammatory response of pancreatic and pulmonary tissues.
