Objective:To investigate the main kinds of food allergen and Mango's hypersensitiveness probability of patients with anaphylactic diseases in our hospital. To analyze some hypersensitive correlation factors of patients,who positive to Mango. To detect the protein components and Immunologic potency of the extracts of Mango fruit allergens which were make by ourselves.To establish the foundation of making and standardizing the self-restraint extracts of food allergen in this area.Methods:①To detect the food allergens of 1901 patients with anaphylactic diseases by intradermal allergen test.According to the traditionary fruit allergen making method to make various kinds self-restraint extracts of Mango fruit allergens by three methods(normal pressure filtrated fluid,supematant fluid by centrifugate,normal pressure filtrated fluid after centrifuging);②To carry out acute toxicity test for the self-restraint extracts of Mango fruit allergen with Kunming mice;③The protein components of the self-restraint extracts of Mango fruit allergen were detected and analyzed by SDS-PAGE and Coomassie Blue staining;④The serum Lrnmunoglobulin G antibody of guinea in the self-restraint extracts of mango fruit allergen were detected by indirect Dot-ELISA using HRP-SPA as the secondary antibody.Results:①The result of this investigation shows that the higher positive allergic rates of food allergen groups in 1901 patients with anaphylactic disease were fresh water fish(65.4%),thalassic-fish(62.5%),shrimp(55.5%),duck(48.5%),mutton(47.3 %),and the positive allergic rate of Mango was 42.3%,it was the seventh.There was a significant difference between the patients who allergic to Mango and their ages or visit time(P<0.001).The positive allergic rates of male:female was 1:1.61,there was a significant difference between them(P<0.05).The main diagnosis were urticaria and contact dermatitis for Mango positive allergic patients,there was statistical significance between positive results of intradermal allergen test and clinical diagnosis(P=0.007),but there was no obviously difference with lesion site(P>0.05);②The highest protein concentration of self-restraint extracts of Mango fruit allergen was made from normal pressure filtrated fluid after centrifuging.To carry out acute toxicity test of Kunming mice with these kinds of allergen,and to observe fourteen days,there is any mice died;③The self-restraint extracts of Mango fruit allergen showed there were nine bands with molecular weight between 30~99 kDa.Three of them were main bands whose molecular weight were 55.6 kDa,47.5 kDa and 32kDa respectively,and each band was clear;④Dot-ELISA results showed that the serum IgG antibody of guinea-pig could react with the self-restraint extracts of Mango fruit antigens,the titer of antibody is 1:64.Immunologic competence was favorable.Conclusion:Mango is one possibly kind of main food allergens in Kunming,it's positive allergic rate was 42.3%in 1901 patients with anaphylactic diseases.There were relations between patients who suffers from allergy to Mango fruit allergen and their ages,sexes,visiting time and clinical manifestation.The highest protein concentration of self-restraint extracts of Mango fruit allergen was made from normal pressure filtrated fluid after centrifuging,we are the first to find that the highest protein concentration of self-restraint extracts of Mango fruit allergen could be made from normal pressure filtrated fluid after centrifuging.These self-restraint extracts of Mango fruit allergen are nontoxic.To get the allergic protein components of the self-restraint extracts of Mango fruit allergen.We are the first to confirm that the serum IgG antibody of guinea-pig could react with the self-restraint extracts of Mango fruit antigens.
