OBJECTIVE:To investigate the expression level of Apelin in the plasma and placenta of healthy pregnant women and gestational diabetic women as well as the relationship between Apelin expression level,gestational blood glucose level and change of the body weight.METHORD:The fasting plasma and placenta samples were collected from 34 full term pregnant women.The fasting plasma Apelin levels were assayed by ELISA and the expression of Apelin in placenta samples were measured by Immunohistochemistry.Register the fasting and postprandial blood glucose levels before delivery,the body weight and body mass index before gestation,body weight before delivery and the weight gain during pregnancy.The relationship between plasma Apelin levels,placenta expression levels of Apelin and other registed indexes were analyzed by statistical software SPSS10.0.RESULT:(1)Weight gain during pregnancy,body weight before delivery and Postprandial blood glucose in GDM women were significantly increased compared with controls(P<0.05).(2) Fasting plasma Apelin level and expression level of Apelin in placenta samples were higher in GDM women than that of controls(P<0.05).(3)The fasting and postprandial blood glucose levels were positively correlated with weight gain during pregnancy(R=0.578,P=0.030;R=0.612,P=0.020).(4)There was not significant correlation between fasting and postprandial blood glucose levels and fasting plasma Apelin(R=0.503,P=0.067;R=0.492,P=0.072).(5) The plasma Apelin was positively correlated with the expression levels of Apelin in the placenta and the weight gain during pregnancy(R=0.656, P=0.011;R=0.837,P=0.000;R=0.807,P=0.000).(6)The expression levels of Apelin in the placenta were positively correlated with postprandial blood glucose,but not correlated with fasting blood glucose.CONCLUSION:The changes of Apelin levels are associated with weight gain during pregnancy and gestational blood glucose,and it may contribute to the pathogenesis of gestational diabetic mellitus.
