Objective To investigate the preventive effect of allogeneic bone marrow hematopoietic stem cell transplantion in NOD mice.Methods Our experiment established a NOD mice allogeneic transplantation model,donors were C57BL/6 mice,recipients were NOD mice. Twenty-one NOD mice(six weeks old)were randomized to two groups of control group and test group.Test group received total body irradiation(TBI)9.5Gy.Four hours after TBI,test group was injected with donor bone marrow cells via tail vein.Control group did not receive any intervention. The blood glucose was examined once a week.At the age of 30 weeks or at the age of the onset of diabetes,all the mice were killed to observe pancreatic histopathology and the incidence of diabetes. Results Compared with control group,the incidence rate of Type 1 DM in NOD mice of test group was lower(Control group:20%;Test group 72.7%;P<0.05),and onset of DM was more postponed(Control group:19.12±5.19w; Test group:27.50±0.71w;P<0.05),The insulitis score decreased markedly (1.02±0.98VS2.26±1.21,P<0.01)in test group than control group,and the severity of insulitis in test group was lower than that in control group(P<0.01).Conclusion It proves the preventive effects of allogeneic bone marrow hematopoietic stem cell transplantion on the onset of type 1 diabetes in NOD mice.
