Objective: The purposes of the experiments were to investigate the effect of bone marrow hematopoiesis in allogeneic mice after Intra-bone marrow bone marrow transplantation(IBM-BMT) and related mechanism in IBM-BMT mice.Methods: 120 recipient BAB L/C (H-2~d) mice were divided randomly into three groups and four batches;each group of each batch had 10 mice. Kill donor mice C57BL/6 (H-2~b) to get bone marrow cells(BMC); adjust BMC density with RPMI1640 to 1 × 10~8/ml (for IV-BMT) and 1 × 10~9/ml (for IBM-BMT) .All recipient mice were lethally irradiated(total dose was 8.5Gy) 5 hours before bone marrow transplantation(BMT).Group A(IBM-BMT),directly inject 30 μl BMC of donor mice into the tibia bone marrow of recipient mice with a microsyringe; Group B(traditional bone marrow transplation,IV-BMT), inject 0.3ml BMC of donor mice into the recipient mice via the tail vein . Group C(blank group), not BMT after pre-treatment.General condition of recipient mice was observed and recorded at 9 A.M. every morning after BMT.The following parameters of recipient mice were observed in the day 1,7,14 post-transplantation : peripheral blood cells,bone marrow mono-nuclear cells (BMMNC) .CFU-S in recipient bone marrow was observed in the day 7,14 post-transplantation.Results: The levels of peripheral WBC,RBC,BPC and the number of BMMNC and CFU-S in recipient bone marrow at the day 7 and 14 in the IBM-BMT group were all significantly higher than those in traditional bone marrow transplation(IV-BMT) group (P<0.01) . The survival rat at 30 days was 20% in IV-BMT group;90% mice of IV-BMT group died of graft versus host diseases(GVHD).All mice in IBM-BMT group survived more than 60 days, without GVHD symptoms. The mice in blank group all died of hematopoiesis failure and the peaking time of deaths were aroud the tenth day afer pre-treatment.Conclusion: Compared with traditional bone marrow transplation (IV-BMT), IBM-BMT further improve the bone marrow hematopoiesis of mice in the hematopoietic re-establishing stage after allo-BMT=...
