Objectives: Research on the expression of rat N-Shc protein; the changes of N-Shc protein phosphorylation of hippocampus in rats after stimulated by H2O2 at different time points; the phosphorylated level of N-Shc protein and PKC5 protein of rat hippocampus and changes of the combination of both after stimulated by H2O2 in different concentrations. To explore the effects of oxidative stress on the function of N-Shc protein of hippocampus in rat. Methods: Western blot technique to detect:①The expression of She family proteins in rat nervous system and other systems;②The expression of N-Shc and PKC8 proteins in brains of different month-old rats;③The changes of hippocampal N-Shc protein phosphorylation after stimulated by H2O2 in different time points;④The changes of phosphorylation level of hippocampal N-Shc protein and PKC8 protein and the combination of both after stimulated by H2O2 in different concentrations.Correcting the collected Data with the density of normal control as the standard, then ussing SPSS for windows 13.0 software for one-way analysis of variances. Results: The expression of rat N-Shc protein concentrated in the nervous system, almost no expression in heart, liver and muscle tissues; only a small amount of expression of brain N-Shc protein in pregnant 12.5 days and 15.5 days, and the expression increased significantly after birth, but PKCδprotein express in the brain of rats in every stages; phosphorylation level of N-Shc protein of hippocampus in rat rising rapidly in 5 min and continuing for 30 min after H2O2 stimulating; Phosphorylation levels of N-Shc protein and PKCδprotein increased gradually after stimulated by H2O2 in different Concentration, there is significant difference between group stimulated by 5 mM H2O2 and the control group (P<0.01); the combination between N-Shc protein and PKCδprotein increased significantly after stimulated by H2O2 in different concentrations, combining weight increasing with the increasion of the concentration of H2O2, there is significant difference between group stimulated by 5 mM H2O2 and the control group (P <0.05). Conclusion: The N-Shc protein in the hippocampus of rats are Phosphorylated when stimulated by H2O2,and the changes of phosphorylation depend on stimulating time and stimulating concentration of H2O2; The stimulation of H2O2 can promote combination between N-Shc protein and PKC8 protein, and the combination increases with the increasing of H2O2 concentration. Our findings suggest that N-Shc protein metergasis under oxidative stress may has a positive role in inhibiting apoptosis.
