Objective:To observe the influence of erdosteine on the activities of superoxide dismutase (SOD), the level of malondialdehyde (MDA) and MMP-9in seizure rats’hippocampus after seizure kindling induced by pentylenetertrazol(PTZ).Method:1. Healthy adult male SD rats were randomly divided into3groups:control,PTZ and erdosteine intervention group.2. Based on the Diehl’s seizure kindling process and its improvement, produce seizure kindling model by PTZ to PTZ and erdosteine group, and gave erdosteine via stomach for pre-disposal treatment before kindling for erdosteine group.3. Detected the activities of SOD and the level of MDA by chemical colorimetric assay,Observed the hippocampal neurons’ damages by hematoxylin-eosinstaining (HE); Observed the distribution of MMP-9positive cells by immunohistochemical staining.Results:1. The activities of SOD:compare to control group, in both PTZ and erdosteine group were low, and PTZ group was significantly more lower(p<0.01). MDA level:in the PTZ group was higher than control group(p<0.01); erdosteine group was lower than PTZ group(p<0.01).2. The results of HE staining:Neurons in hippocampus of the control group were normal microscopically. The PTZ group which has scattered arrangement of hippocampal neurons, reduction in the number, shrunken of cell body, red staining cytoplasm and condensated nuclear appeared a large number of acidophilic neurons. There were less addophilic neurons in the hippocampus of erdosteine group. There were significant differences between than (p<0.01).3. MMP-9positive cells:The staining intensity in nonnal group and erdosteine intervention group significantly more decreased than in PTZ group (p<0.05).Conclusion:Oxidative stress, cytokines diversification and neuronal damage go with seizure, erdosteine could inhibit neuronal damage, scavenge free radicals, and might inhibit the activation of MMP-9. It has protective effect on brain.
