Objective to evaluate the clinical response rate and the toxicity of neoadjuvant chemotherapy with capecitabine and oxaliplatin for advanced gastric cancer,and analyzed its efficacy and safety.Methods Patients with preoperative histologically confirmed,baseline stageâ…¢orâ…£gastric carcinoma are enrolled in this study.Patients will receive three-weekly cycle chemotherapy of capecitabine at 1000mg/m~2 orally twice a day on days1 to 14,oxaliplatin at 130mg/m~2 intravenously on day1.After third cycle of chemotherapy,surgery was attempted.Results from 2004 to 2008,49 patients are enrolled in this study,in which 47 patients completed 3 cycles of chemotherapy and were available for clinical response evaluation.The clinical response was 46.8%and 3 patients achieved clinical complete response.30 patients received surgery,29 of 30 patients received curative resection and 23 patients (67.6%)reached R0 resection.The major nonhemological toxicities were nausea/vomiting and hand-foot syndrome;the major hemological toxicities were anemia and leukocytopenia.Most adverse events were in gradeâ… /â…¡in severity.The median follow-up time was 23 months.1-year,2-year survival was 83%,59.7%and R0 resection was significantly related with the overall survival.Conclusion Neoadjuvant chemotherapy with capecitabine was an effective and well-tolerated regimen for advanced gastric cancer.
